摘要
【目的】分析从内蒙古红花尔基、河北丰宁、河北围场以及辽宁章古台地区收集的樟子松(Pinus sylvestris var.mongolica)优树资源遗传多样性及其遗传结构差异,构建樟子松优良单株的指纹图谱,为优树资源保存和利用提供依据。【方法】利用简单重复序列(simple sequence repeats,SSR)分子标记技术,根据所得结果进行遗传多样性分析,通过非加权组平均法(UPGMA)进行基于亲缘关系的聚类分析,构建99个樟子松优树指纹图谱。【结果】12对SSR引物共扩增35个等位基因,每个位点平均扩增出5.25个等位基因和3.112个有效等位基因,不同位点的PIC值在0.202~0.932之间,平均PIC值为0.558;聚类分析表明,99个樟子松优良单株中相同来源的个体并没有聚为一类,各单株的聚类与现有栽培区不存在明显的规律,樟子松优树资源遗传组成复杂;指纹图谱的构建结果表明,引物lw_isotig17679、lw_isotig21953、lw_isotig27940、lw_isotig02842、lw_isotig00080、lw_isotig05123、lw_isotig02138进行组合,可鉴别99份樟子松优良单株材料,分辨率为100%。【结论】4个地区的樟子松优树资源遗传多样性丰富,个体和群体的遗传距离分散均匀,樟子松优良单株指纹图谱能够用于樟子松个体鉴定和亲缘关系分析。
【Objective】Understand the genetic diversity and to construct the fingerprints of superior individual trees of Pinus sylvestris var.mongolica from Honghuaerji,Fengning,Weichang and Zhanggutai in order to provide a basis for the preservation and utilization of excellent tree resources.【Method】Genetic diversity was analyzed according to the results by simple sequence repeats molecular marker method,the unweighted group average(UPGMA)method was used for cluster analysis based on genetic relationship and construction of fingerprints of 99 superior individuals.【Result】Total of 35 alleles were identified using 12 pairs of SSR primers,with a mean of 5.25 alleles and 3.112 effective alleles per locus.PIC values of different loci ranged from 0.202 to 0.932,with an average of 0.558;The results of cluster analysis showed that individuals from the same origin of 99 materials were not clustered into one group according to their genetic relationship.There were not obvious correlations between grouping and current growing regions of the clones.The results of fingerprinting showed that the combination of primers lw_isotig17679,lw_isotig21953,lw_isotig27940,lw_isotig02842,lw_isotig00080,lw_isotig05123 and lw_isotig02138 could identify 99 superior materials,the resolution was 100%.【Conclusion】The genetic diversity of Pinus sylvestris var.mongolica in four areas is relatively high.The fingerprint of superior tree can be used to identify the individual and genetic relationship analysis.
