甘肃省珍稀野生食用菌新记录种——台湾块菌

A new record species of rare edible fungus in Gansu Province——Tuber formosanum

【目的】对采自甘肃省陇南市康县的块菌标本通过形态特征和分子标记进行物种鉴定。【方法】对块菌标本进行宏观和显微形态的研究;对其ITS序列进行扩增和测序,构建分子系统发育树,解析其系统发育位置。【结果】系统发育树显示,标本(zhu4361-1、zhu4361-2、zhu4361-3、zhu4361-4)以100%的支持率与台湾块菌的相关序列聚于一支,结合形态学研究结果,最终确定所采集块菌标本均为台湾块菌(Tuber formosanum H.T.Hu&Y.Wang.),凭证标本保存于西北师范大学生命科学学院标本室。【结论】采自甘肃省陇南市康县的块菌标本为台湾块菌,是甘肃省的新记录种。

【Objective】To identify the truffle specimens collected from Kangxian Country,Longnan City,Gansu Province.【Method】The morphology of the truffle specimens were analyzed with macroscopic and microscopic examination,ITS sequences were obtained by PCR and sequencing,and the molecular phylogenetic tree was constructed based on ITS to clarify the phylogenetic position.【Result】The molecular phylogenetic tree showed that the specimens(zhu4361-1,zhu4361-2,zhu4361-3,zhu4361-4)were united with sequences of Tuber formosanum H.T.Hu&Y.Wang in a bootstrap value of 100%,and were identified as Tuber formosanum based on both morphological and molecular phylogenetic evidence.The voucher specimens were preserved in the herbarium of Northwest Normal University.【Conclusion】The truffle specimens collected from Kang Country,Longnan City,Gansu Province is Tuber formosanum,which is a newly recorded species in Gansu Province.
