麻黄升麻汤对急性肺损伤大鼠微生物多样性和碳水化合物酶谱的影响 被引量:3

Effect of Mahuang Shengma Decoction on Microbial Diversity and Carbohydrate Enzyme Spectrum in Rat Model of Acute Lung Injury

摘要 目的基于宏基因组学方法探讨麻黄升麻汤对肠缺血-再灌注诱发急性肺损伤模型大鼠微生物组成及碳水化合物酶类(CAZy)功能谱的影响。方法Wistar雄性大鼠32只,随机分为假手术组、模型对照组、麻黄升麻汤组(17 g·kg^(-1))、地塞米松组(5 mg·kg^(-1)),每组8只,治疗组均给予灌胃给药,检测大鼠肺泡灌洗液中巨噬细胞浸润情况,苏木精-伊红(HE)染色观察大鼠肺、肠组织形态变化。采集结肠内容物,提取DNA,进行宏基因组分析。结果麻黄升麻汤组大鼠细支气管及肺泡扩张充血改善,肺泡巨噬细胞浸润减轻;宏基因组学分析表明麻黄升麻汤组普雷沃菌属、拟杆菌属、乳杆菌属、瘤胃球菌属丰度较模型对照组升高,并能激活和促进CAZy的利用,从而降解和消化中药中植物细胞壁多糖成分。结论麻黄升麻汤可抑制肠缺血-再灌注过度炎症反应,从“药物微生物组学”角度出发,其作用机制可能与提高特定肠道微生物中CAZy的丰度或葡萄糖苷酶活性,增强中药活性成分的生物利用度有关。 Objective To investigate the effects of Mahuang Shengma decoction on microbial composition and carbohydrate enzyme(CAZy)function spectrum in rat model of acute lung injury induced by intestinal ischemia-reperfusion based on metagenomic method.Methods A total of 32 Wistar rats were divided into sham group,model control group,Mahuang Shengma decoction group and dexamethasone group(n=8 in each group).The infiltration of macrophages in alveolar lavage fluid of rats was detected,and the morphological changes of lung and intestine tissues were observed by HE staining.Colon contents were collected and DNA was extracted for metagenomic analysis.Results Ephedra shengma decoction could improve bronchiole and alveolar dilatation and congestion and alveolar macrophage infiltration.Metagenomic analysis showed that the abundance of Prevotella,Bacteroides,Lactobacillus and Ruminococcus in Mahuang Shengma decoction group was higher than that in model control group,and it could activate and promote the utilization rate of CAZy.In order to degrade and digest the plant cell wall polysaccharide components in Traditional Chinese Medicine.Conclusion From the perspective of pharmacomicrobiome,increasing the abundance of CAZy or glucosidase activity in certain intestinal microorganisms and enhancing the bioavailability of the active components of Traditional Chinese Medicine may be the mechanism of Mahuang Shengma decoction inhibiting the inflammatory response of intestinal ischemia-reperfusion.
