摘要
目的探索应用盐酸洛哌丁胺制备小鼠便秘模型的最佳用药剂量和用药时间。方法选取2个月龄C57BL/6J小鼠50只,雌雄各半。随机分为空白对照组与便秘模型组。便秘模型组根据盐酸洛哌丁胺剂量分为50、60、70、80mg/kg 4个组,将盐酸洛哌丁胺溶于1 mL纯水中,于每天早、晚7:00灌胃给药;空白对照组给予等量纯水,持续7 d,观察小鼠每日体重、大便量和性状、活动状态、毛发状态、存活情况,灌胃7 d时同时灌胃墨水,观察肠推进率。通过小鼠粪便量、性状及肠道推进率的变化判断小鼠便秘模型是否制备成功,并确定制备便秘模型的最佳用药剂量和时间。结果与空白对照组相比,盐酸洛哌丁胺各剂量组小鼠的粪便量和性状、小肠推进率差异均有统计学意义(P<0.05);与50、60及80 mg/kg组比较,70 mg/kg组盐酸洛哌丁胺灌胃7 d制备的小鼠便秘模型效果好,制备的便秘模型较稳定。结论70 mg/kg盐酸洛哌丁胺连续灌胃7 d,每天早晚各1次,可制备稳定的小鼠便秘模型。
Objective To explore the optimal dosage and time of loperamide hydrochloride in the preparation of mouse constipation model.Methods Two month old C57 BL/6 J mice were randomly divided into the blank control group and the loperamide hydrochloride 50 mg/kg,60 mg/kg,70 mg/kg and 80 mg/kg groups.Loperamide hydrochloride was dissolved in 1 mL pure water and administered by gavage at 7:00 and 19:00 every day.The mice in the blank control group were given the same amount of pure water for 7 days.The daily weight,fecal volume and characteristics,intestinal propulsive rate,activity status,hair status and survival status of mice were observed.The successful preparation of constipation model was judged by fecal volume,characteristics of mice and intestinal propulsion rate.The optimal dosage and time were determined.Results Compared with the blank control group,there were significant differences in the amount and character of feces and intestinal propulsion rate of mice in each dose of loperamid hydrochloride groups(P<0.05).Compared with 50,60 and 80 mg/kg groups,the constipation model of mice prepared by intragastric administration of 70 mg/kg loperamide hydrochloride for 7 days had the best effect.Conclusion A stable model of constipation in mice can be established by continuous intragastric administration of 70 mg/kg loperidamide hydrochloride twice a day for 7 days.
