摘要
采用HPLC法测定新疆小枝玫瑰花中金丝桃苷的含量.色谱条件为:Hypersil ODS色谱柱(250mm×4.6 mm,5μm),柱温35℃,流动相为乙腈-0.2%磷酸,梯度洗脱,流速1.0mL·min^(-1),检测波长360nm,进样量20μL.结果表明,金丝桃苷浓度在0.0147~0.0735mg·mL^(-1)范围内与峰面积线性关系良好;平均加标回收率为101.15%,RSD值为1.59%;新疆小枝玫瑰花中金丝桃苷含量为0.393mg·g^(-1).该方法准确可靠,可用于新疆小枝玫瑰花中金丝桃苷含量的测定.
We determined the content of hyperoside in branchlet rose flowers from Xinjiang by adopting the HPLC method.The chromatographic conditions are determined as follows:the analysis is performed on Hypersil ODS column(250 mm×4.6 mm,5μm),the column temperature is set to be 35℃,acetonitrile-0.2%phosphoric acid is adopted as the mobile phase for gradient elution,and the flow rate of 1.0 mL·min^(-1),the detection wavelength of 360 nm,and the sample volume of 20μL are adopted.The results show that the concentration of hyperoside bears a good linear relationship with the peak area within the range of 0.0147-0.0735 mg·mL^(-1).The average recovery is 101.15%with the RSD value of 1.59%,and the content of hyperoside in branchlet rose flowers from Xinjiang is 0.393 mg·g^(-1).This method is accurate and reliable,which can be used for the determination of hyperoside in branchlet rose flowers from Xinjiang.
