基于生信分析DPP7在结肠癌中的诊断及预后价值

目的本文旨在对DPP7基因在结肠癌患者中潜在的诊断和预后意义进行探讨。方法从TCGA公共数据库中下载结肠癌的相关样本资料,过滤得出包含一般临床信息、分期、预后等相关临床信息的样本资料。首先分析非配对样本中结肠癌组织与正常组织中DPP7基因的表达值之间有无差异,再进行筛选得出包括癌与癌旁组织的结肠癌配对样本,分析配对样本中癌与癌旁组织中DPP7的基因表达有无差异。基于DPP7的基因表达构建出预测结肠癌组织或是正常组织的ROC曲线。比较结肠癌不同肿瘤T、N、M分期中DPP7的表达差别。根据DPP7的基因表达值高低将结肠癌样本分为DPP7高表达组与DPP7低表达组,分析DPP7的高表达与低表达对结肠癌生存预后指标OS、DSS及PFI的影响。最后对DPP7与结肠癌中24种免疫细胞相关性进行分析。结果无论是非配对样本还是配对样本,DPP7在结肠癌中的基因表达值均较正常组织的表达值显著升高,并且具有统计学差异。通过DPP7在结肠癌中的表达情况构建ROC曲线,其AUC及CI值显示DPP7对结肠癌具有很好的预测作用。DPP7在结肠癌不同肿瘤N、M分期中的表达具有显著差异,且具有统计学意义。存在低表达DPP7的患者具有较好的OS、DSS和PFI。DPP7的表达与结肠癌中多种免疫细胞具有相关性。结论DPP7有可能成为结肠癌的诊断及预后指标,DPP7与结肠癌中的免疫细胞密切相关,与免疫浸润有关。

Objective To explore the prognostic and diagnostic value of DPP7 in colon cancer.Methods Download sample data about colon cancer from the TCGA database,After filtering,we obtained data including general clinical information,stage,prognosis,and other clinical information.We first analyzed the difference of DPP7 expression in colon cancer tissue and normal tissue of unpaired samples,then screened paired samples including colon cancer tissue and normal adjacent tis⁃sue,analyzed the DPP7 expression in the paired samples.Constructed ROC curve based on DPP7 gene-expression,com⁃pared the difference of DPP7 in different tumor stages,analyzed the prognosis of colon cancer by high and low expression of DPP7 Influence,analyzed the correlation between DPP7 and 24 immune cells.Results The gene expression of DPP7 in colon cancer was statistically significantly higher than that of normal tissues both for pairing and unpairing samples.ROC curve shows that DPP7 has a good predictive effect on colon cancer.The expression of DPP7 in different N and M stages of colon cancer is significantly different,and patients with lower expression of DPP7 have longer OS,DSS and PFI.The expression of DPP7 is correlated with a variety of immune cells in colon cancer.Conclusion DPP7 may be a diagnostic and prognostic in⁃dicator of colon cancer.DPP7 is closely related to immune cells and immune infiltration of colon cancer.
