初探eDNA在贵州草海入侵种克氏原螯虾监测中的应用 被引量:2

摘要 近年来克氏原螯虾入侵草海,对草海生态系统中的水生植物、两栖动物和无脊椎动物的生存造成威胁,严重影响入侵地生物的多样性。为进一步分析克氏原螯虾的引入或扩散初期的监测、预警和应急处理提供有效依据,研究了环境DNA技术在监测草海克氏原螯虾分布应用中的最适条件。结果表明,4℃保存8h内,0.45μm孔径硝酸纤维膜过滤2000ml水体提取eDNA效果最好,且过滤水体体积越大,富集eDNA浓度越高。越靠近城市人类活动较活跃的采样点,克氏原螯虾分布密度越高,表明人类活动可能是导致克氏原螯虾成功入侵草海的主要原因。 In recent years,that the invasion of Procambarus clarkii in Caohai has caoused a serious threat to the survival of aquatic plants,amphibians and invertebrates in the Caohai ecosystem,and has a serious impact on the biodiversity of the invasion area.This study explored the optimal conditions of eDNA(environmental DNA)technology in monitoring the distributin ofP.clarkii.The results showed that the optimal eDNA extraction effect of 2000 mL water was filtered with 0.45μm nitrocellulose membrane after storage at 4℃for 8 hours,and the larger the volume of filtered water,the higher the concentration of enriched eDNA.In addition,the distribution density ofP.charkii is hisher at sampling points close to the city where human activities are more active,indicating that human activities may be the main reason for the successful invasion of P.clarki into Caohai.This study provides an effective basis for further analysis of the monitoring,early warning and emergency treatment of the introduction or spread of P.clarkii.
