保健食品中叶黄素总量的高效液相色谱分析测定

Determination of lutein in health food by HPLC

建立了一种保健食品中叶黄素总量的高效液相色谱分析方法。采用乙醇溶液分散溶解,在碱性溶液中皂化成游离叶黄素,再用有机溶剂萃取,0.1%BHT乙醇溶液定容,经C18色谱柱分离,采用高效液相色谱法外标法定量。结果表明,在4μg/mL~50μg/mL的范围内线性良好,得到回归方程为y=95570.5x-164969,相关系数为0.9994,精密度RSD为0.33%,重复性RSD为1.67%,加标回收率在102.10%~103.91%之间。该方法准确可靠,能用于保健食品中叶黄素总量的测定。

A method of High-performance liquid chromatography has been developed for the determination of Lutein in dietary supplement.Samples were dissolved in ethanol,saponified in alkaline solution,and then free Lutein was extracted by organic solvent,fixed by 0.1%BHT ethanol solution,separated by C18 column,and quantified by external standard method of High-performance liquid chromatography.The regression equation was y=95570.5x 164969,the linearity was good in the range of 4μg/mL~50μg/mL,the correlation Coefficient was 0.9994,the RSD of precision was 0.33%,the RSD of repeatability was 1.67%,and the recoveries were between 102.10%~103.91%.The method is accurate and reliable,and can be used for the determination of lutein in health food.
