摘要
目的探讨环状RNA hsa_circ_0009244(circ-9244)在食管鳞癌组织中的表达及其与食管鳞癌患者预后的关系,分析下调circ-9244对食管鳞癌细胞增殖、迁移及侵袭的影响。方法运用实时荧光定量聚合酶链反应检测circ-9244在食管鳞癌细胞及正常食管上皮细胞Het-1A中的表达水平,构建circ-9244下敲质粒PLKO.1-circ-9244-shRNA并转染至高表达circ-9244的食管鳞癌细胞KYSE410、KYSE520中,运用MTS实验、划痕实验及Transwell实验检测其下调后对食管鳞癌细胞增殖、迁移和侵袭的影响。结果实验组食管鳞癌细胞增殖效率、划痕愈合率及穿膜细胞数明显低于空载组,差异均有统计学意义(KYSE410细胞:t=12.345,P<0.001;t=10.910,P<0.001;t=11.040,P<0.001;KYSE520细胞:t=7.941,P<0.001;t=8.499,P<0.001;t=10.720,P<0.001)。结论下调circ-9244可抑制食管鳞癌细胞的增殖、迁移和侵袭能力。
Objective To investigate the expression of circular RNA hsa_circ_0009244(circ-9244)in the esophageal squamous cell carcinoma tissues and its relationship with the prognosis of patients with esophageal squamous cell carcinoma,and to analyze the effects of down-regulation of circ-9244 on the proliferation,migration and invasion of esophageal squamous cancer cells.Methods Quantitative real-time polymerase chain reaction was used to examine the expression of circ-9244 in the esophageal squamous cell carcinoma cell lines and the esophageal epithelial cell Het-1A.Constructed circ-9244 knockdown plasmid PLKO.1-circ-9244-shRNA was transfected into the KYSE410 and KYSE520 of esophageal squamous cancer cells high-expressed circ-9244,and then MTS experiment,wound healing experiment and transwell test were employed to explore the proliferation,migration and invasion of esophageal squamous cell carcinoma cells with down-regulation of circ-9244.Results The cell proliferation efficiency,scratch healing rate and the number of perforated cells in the experimental group were significantly lower than those in the empty vector group,the differences were statistically significant(KYSE410 cells:t=12.345,P<0.001;t=10.910,P<0.001;t=11.040,P<0.001;KYSE520 cells:t=7.941,P<0.001;t=8.499,P<0.001;t=10.720,P<0.001).Conclusion Down-regulation of circ-9244 inhibited the proliferation,migration and invasion of esophageal squamous carcinoma cells.
