摘要
采用硅胶柱色谱、Sephadex LH-20凝胶柱色谱和反相高效液相制备色谱(HPLC),从黄连内生真菌Albifimbria viridis固体发酵物中分离纯化得到8个化合物,通过波谱学数据分析并结合其物理性质,将其鉴定为:Roridin J(1)、verrucarin A(2)、verrucarin L acetate(3)、Calcarisporin B1(4)、verrucarin J(5)、Isotrichoverrin A(6)、Isotrichoverrin B(7)、16-hydroxyroridin A(8).以上化合物均首次从Albifimbria viridis中分离得到.利用MTT法检测了化合物1~8对MCF-7和A549细胞的体外细胞毒活性,结果显示化合物4、6、8对MCF-7(人乳腺细胞)具有显著的抑制活性.
By using gel column chromatography,sephadex LH-20,and ODS-HPLC,eight compounds were isolated from the endophytic fungus Albifimbria viridis of Coptidis chinensis.The structures of the compounds were determined to be Roridin J(1),verrucarin A(2),verrucarin L acetate(3),Calcarisporin B1(4),verrucarin J(5),Isotrichoverrin B(6),Isotrichoverrin B(7),16-hydroxyroridin(8).All compounds were isolated from the fermentation of Albifimbria viridis for the first time.The cytotoxicity of compounds 1-8 on MCF-7 and A549 cells in vitro were detected by MTT assay.The results showed that compounds 4,6 and 8 had significant inhibitory activity on MCF-7(human breast cells).
