摘要
目的:研究柴胡皂苷(SS)对离体大鼠缺血-再灌注(I/R)的心肌保护作用。方法:健康SD大鼠30只被随机均分为5组:空白对照组、SS组(空白对照组基础上加用SS)、I/R组、SS+I/R组(I/R组基础上加用SS)、SS+3-甲基腺嘌呤(3-MA)+I/R组(SS+I/R组基础上加用3-MA)。比较各组梗死心肌范围、心脏做功指标、冠脉流出液心肌酶活性、心肌组织氧化应激指标及心肌组织微管相关蛋白轻链3(LC3)的表达。结果:与SS组比较,再灌注2h末,SS+I/R组心脏做功显著降低;SS+3-MA+I/R组心脏做功显著升高;I/R组dp/dt_(max)及左室内压(LVDP)恢复率、GSH-px、CuZn-SOD水平显著下降,心肌梗死面积、MDA水平、心肌酶活性、LC3表达水平显著升高,P<0.05或<0.01;与I/R组比较,SS+I/R组dp/dt_(max)及LVDP恢复率、GSH-px及CuZn-SOD水平显著升高,心肌梗死面积、MDA水平及心肌酶活性显著降低;SS+3-MA+I/R组dp/dt_(max)及LVDP恢复率、GSH-px及CuZn-SOD水平显著降低,心肌梗死面积、MDA水平及心肌酶活性显著升高,P<0.05或<0.01。结论:柴胡皂苷对离体心脏缺血-再灌注损伤具有保护作用。
Objective:To study myocardium protection effect of Saikosaponin(SS)on ischemia reperfusion(I/R)in isolated rats.Methods:A total of 30 healthy SD rats were randomly and equally divided into 5 groups:blank control group,SS group(received SS based on blank control group),I/R group,SS+I/R group(received SS based on I/R group)and SS+3-methyladenine(3-MA)+I/R group(received 3-MA based on SS+I/R group).Range of infarcted myocardium,cardiac work indexes,myocardial enzyme in coronary outflow fluid,myocardial oxidative stress index and myocardial tissue microtubule-associated protein light chain 3(LC3)were compared among all groups.Results:Compared with SS group,2 h after reperfusion,there were significant reductions in cardiac work in SS+I/R group,and there were significant rise in cardiac work in SS+3-MA+I/R group;there were significant reductions in recovery rate of dp/dtmax and left ventricular developed pressure(LVDP),levels of GSH-px and CuZn-SOD,and significant rise in myocardial infarction area,level of MDA,myocardial enzyme activities and LC3 protein expression in I/R group,P<0.05 or<0.01.Compared with I/R group,there were significant rise in recovery rate of dp/dt_(max) and LVDP,levels of GSH-px and CuZn-SOD,and significant reduction in myocardial infarction area,level of MDA and myocardial enzyme activities in SS+I/R group;and there were significant reductions in recovery rate of dp/dt_(max) and LVDP,levels of GSH-px and CuZn-SOD,and significant rise in myocardial infarction area,level of MDA and myocardial enzyme activities in SS+3-MA+I/R group,P<0.05 or<0.01.Conclusion:Saikosaponin possesses myocardium protective effect on ischemia reperfusion injury of isolated rats after ischemia reperfusion.
