摘要
目的 采用网络药理学方法研究黄芩治疗痤疮的作用机制。方法 通过中药系统药理学数据库与分析平台(TCMSP)检索黄芩的活性成分和靶标,采用GeneCards和在线人类孟德尔遗传(OMIM)数据库筛选痤疮的潜在靶点;通过Cytoscape3.7.1软件绘制黄芩治疗痤疮的“成分-靶点-通路-疾病”网络图,利用STRING数据库构建蛋白质-蛋白质相互作用(PPI)网络,并对关键靶点进行基因本体(GO)及京都基因与基因组百科全书(KEGG)通路分析。结果 从黄芩中筛选出与痤疮有关的活性成分29个,通过干预36个靶点、247条通路治疗痤疮;富集分析显著的生物功能主要为核受体活性、转录因子活性,直接配体调节的序列特异性DNA结合、类固醇激素受体活性、RNA聚合酶Ⅱ转录因子结合、四吡咯结合等。KEGG信号通路结果显示,黄芩治疗痤疮的关键基因显著富集在白细胞介素-7(IL-7)信号通路、肿瘤坏死因子(TNF)信号通路、缺氧诱导因子-1(HIF-1)信号通路、松弛素信号通路、磷脂酰肌醇-3激酶蛋白激酶B(PI3K-Akt)信号通路等。结论 黄芩主要通过多成分、多靶点、多通路的作用治疗痤疮,而非通过单一机制发挥作用。
Objective To study the mechanism of Scutellariae Radixin the treatment of acne based on network pharmacology.Methods The active components and targets of Scutellariae Radixwere searched through Traditional Chinese Medicine Systems Pharmacology(TCMSP) database and analysis platform,Gene Cards and Online Mendelian Inheritance In Man(OMIM) database were used to screen the potential targets of acne;The “component-target-pathway-disease” network diagram of Scutellariae Radixin the treatment of acne was drawn through Cytoscape 3.7.1 sof tware,the protein-protein interaction(PPI) network was constructed by using the STRING database,and Gene Ontology(GO) enrichment analysis and Kyoto Enyclopedia for genes and genomes(KEGG) signal pathway enrichment analysis were carned out for key targets.Results Twenty-nine active components related to acne were selected from Scutellariae Radix,and acne was treated by intervening 36 targets and 247 pathways;The significant biological functions of GO enrichment analysis were mainly nuclear receptor activity,transcription factor activity,sequence specific DNA binding regulated by direct ligand,steroid hormone receptor activity,RNA polymerase Ⅱ transcription factor binding and tetrapyrrole binding etc. The results of KEGG signal pathway showed that the key genes of Scutellariae Radixfor the treatment of acne were significantly enriched in interleukin-7(IL-7) signal pathway,tumor necrosis factor(TNF) signal pathway,hypoxia inducible factor-1(HIF-1) signal pathway,relaxin signal pathway,phosphatidylinositol-3 kinase(PI3K-Akt) signal pathway and so on.Conclusion Scutellariae Radix mainly treats acne through multi-component,multi-target and multi-path effects,rather than through a single mechanism.
