摘要
目的研究热习服(HA)对热应激(HS)诱导的PC12和H9C2细胞存活率、炎性因子及氧化应激的影响,探讨HA对HS的防护效应,建立基于PC12和H9C2热习服细胞模型。方法将PC12和H9C2细胞分为:对照组(C组,37℃)、HA组(39.5℃,3 h/d,连续3 d)、PC12细胞HS组(43℃、2 h)、H9C2细胞HS组(44℃、2 h)、HA+HS组(细胞经HA后,随后进行HS)。利用CCK-8法检测各组两种细胞的存活率,酶联免疫吸附法(ELISA)检测炎性因子及氧化应激水平。结果(1)HA可显著缓解HS诱导的细胞存活率的降低;(2)HA可降低HS诱导的细胞炎性反应:提高白细胞介素-4(IL-4)水平,降低肿瘤坏死因子-α(TNF-α)、IL-1β、IL-6和热休克蛋白70(HSP70)水平(P<0.05);(3)HA可提高HS诱导的细胞抗氧化能力:提高谷胱甘肽过氧化物酶(GSH-Px)水平、降低超氧化物歧化酶(SOD)和丙二醛(MDA)水平(P<0.05)。结论HA可减轻由HS导致的细胞炎性反应和氧化应激损伤,提高PC12和H9C2细胞的耐热能力,具有HA防护效应。
Objective To investigate the effects of heat acclimation(HA)on cell viability,inflammatory factors and oxidative stress response induced by heat stress(HS)in PC12 and H9C2 cells,explore the protective effects of HA on HS,and to construct HA and HS cell models based on PC12 and H9C2.Methods PC12 and H9C2 cells were divided into 4 groups:C group(37℃),HA group(39.5℃,3 h/d for 3 d),HS group of PC12 cells(43℃,2 h),HS group of H9C2 cells(44℃,2 h),and HA+HS group(cells were subjected to HA followed by HS).Cell vitality was detected using a cell counting kit 8(CCK-8),and the enzyme-linked immunosorbent assay(ELISA)was used to determine the inflammatory factors and oxidative stress levels.Results(1)HA could significantly mitigate the decline of HS-induced cell survival rate.(2)HA could decrease the HS-induced cellular inflammation response.IL-4 levels increased while TNF-α,IL-1β,IL-6 and HSP70 levels decreased(P<0.05).(3)HA could increase the HS-induced cellular antioxidant capacity in that GSH-Px levels were increased while SOD and MDA levels were decreased(P<0.05).Conclusion HA can reduce cellular inflammation response and oxidative stress damage caused by HS,while improving the heat tolerance of the PC12 and H9C2 cells.HA may protect cells from HS to some extent.
