摘要
目的利用表面多孔型的色谱柱对鱼腥草的特征图谱和含量测定方法进行快速分析,在常规液相色谱仪器系统中,实现高效、快速的中药定性定量分析。方法使用普通高效液相色谱柱(TC-C_(18),4.6 mm×250 mm,5μm)和Poroshell C_(18)色谱柱(4.6 mm×100 mm,2.7μm)分别对鱼腥草配方颗粒进行定性和定量分析。结果鱼腥草配方颗粒特征图谱分析,TC-C_(18)普通色谱柱需要80 min完成分析,Poroshell 120 EC-C_(18)色谱柱仅用32 min即可完成。槲皮苷含量测定分析,TC-C_(18)普通色谱柱要16 min完成分析,Poroshell 120 EC-C_(18)色谱柱仅用4 min即可完成。结论使用粒径为2.7μm的表面多孔型Poroshell色谱柱,可以在常规液相色谱仪上实现鱼腥草配方颗粒的快速液相色谱分析,为其产品产业化检测效率的提高和检测成本的降低提供了依据。
OBJECTIVE To develop rapid method for characteristic chromatogram and assay test of Houttuynia cordata formula granules by poroshell column with superficially porous particles(SPP)on conventional HPLC instrument.METHODS A total porous particles(TPP)HPLC column(TC-C_(18),250 mm×4.6 mm,5μm)and an SPP column(Poroshell 120 EC-C_(18),100 mm×4.6 mm,2.7μm)were utilized to develop the rapid methods for the qualitative and quantitative determination of Houttuynia cordata formula granules respectively.RESULTS The time of characteristic chromatogram on normal TC-C_(18) was 80 min,while on Poroshell 120 EC-C_(18)was shorten to 32 min.The content determination of quercitrin could be finished in 4 min on Poroshell 120 EC-C_(18),while 16 min on normal TC-C_(18).CONCLUSION The characteristic chromatogram and assay of Houttuynia cordata formula granules can be quickly and efficiently tested on the SPP column with a particle size of 2.7μm on a conventional HPLC instrument,which is helpful to improve the detection efficiency and reduce the detection cost in industrial testing.
