摘要
目的:研究平胃散加味对非酒精性脂肪肝(NAFLD)大鼠肝细胞凋亡、炎症介质及氧化应激的影响。方法:选取48只无特定病原体(SPF)级健康SD大鼠,按照随机数字表法分为对照组、模型组、低剂量组和高剂量组,每组12只。采用OpenSource Diets^(TM)高脂饲料喂养12周,制备NAFLD大鼠模型。各组大鼠根据10 mL/kg给药,对照组和模型组大鼠给予等剂量生理盐水灌胃,低剂量大鼠给予1.89 g生药/kg灌胃,高剂量大鼠给予7.56 g生药/kg灌胃,1次/d,连续灌胃8周。比较各组大鼠肝脏指数、肝功能、炎症介质和氧化应激变化。结果:模型组、低剂量组和高剂量组肝脏指数高于对照组(P<0.05);低剂量组和高剂量组肝脏指数低于模型组,且高剂量组低于低剂量组(P<0.05)。模型组、低剂量组和高剂量组血清谷草转氨酶(GOT)和谷丙转氨酶(GPT)水平均高于对照组(均P<0.05);低剂量组和高剂量组血清GOT和GPT水平低于模型组,且高剂量组低于低剂量组(P<0.05)。模型组、低剂量组和高剂量组血清白细胞介素-2(IL-2)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)水平均高于对照组(均P<0.05);低剂量组和高剂量组血清IL-2、IL-6和TNF-α水平低于模型组,且高剂量组低于低剂量组(P<0.05)。模型组、低剂量组和高剂量组血清丙二醛(MDA)水平高于对照组,而超氧化物歧化酶(SOD)水平低于对照组(P<0.05);低剂量组和高剂量组血清MDA水平低于模型组,而SOD水平高于模型组(P<0.05);高剂量组血清MDA水平低于低剂量组,而SOD水平高于低剂量组(P<0.05)。结论:平胃散加味可减轻NAFLD大鼠肝细胞凋亡,减轻肝脏炎症反应及调节肝脏氧化应激。
Objective:To study the effects of modified Pingwei Powder on hepatocyte apoptosis,inflammatory factors and oxidative stress in rats with nonalcoholic fatty liver disease(NAFLD).Methods:A total of 48 healthy SPF SD rats were divided into control group,model group,low-dose group and high-dose group by random number table,with 12 in each group.NAFLD rat model was established by OpenSource DietsTM high-fat diet for 12 weeks.Rats in low-dose and high-dose groups were given 1.89 g crude drug/kg and 7.56 g crude drug/kg(10 mL/kg)by gavage,respectively,and those in the control and model groups were given equal dose of normal saline by gavage,once a day for 8 weeks.The changes of liver index,liver function,inflammatory factors and oxidative stress were compared.Results:The liver index of model,low-dose and high-dose groups was higher than that of control group(P<0.05),and the liver index of low-dose and high-dose groups was lower than that of model group,with high-dose group lower than low-dose group(P<0.05).The serum glutamic oxaloacetic transaminase(GOT),glutamic pyruvic transaminase(GPT),interleukin-2(IL-2),interleukin-6(IL-6)and tumor necrosis factorα(TNF-α)levels of model,low-dose and high-dose groups were higher than those of control group(P<0.05),and the levels of low-dose and high-dose groups were lower than those of model group,with high-dose group lower than low-dose group(P<0.05).The serum malondialdehyde(MDA)level of model,low-dose and high-dose groups was higher while the superoxide dismutase(SOD)level was lower than those of control group(P<0.05),and serum MDA level of low-dose and high-dose groups was lower while the SOD level was higher than thoses of model group(P<0.05),with serum MDA level of high-dose group lower while SOD level higher than those of low-dose group(P<0.05).Conclusion:Modified Pingwei Powder could reduce hepatocyte apoptosis,decrease liver inflammation and regulate liver oxidative stress in NAFLD rats.
