复方野菊花提取物对代谢性高血压模型大鼠的降压作用研究 被引量:5

摘要 目的观察复方野菊花提取物对高糖、高脂、饮酒所致代谢性高血压模型大鼠的作用及可能机制。方法采用高糖、高脂饲料复合梯度饮酒建立代谢性高血压大鼠模型,造模成功后,大鼠每天灌胃给予复方野菊花提取物(5.25、3.50、1.75 g·kg^(-1))连续4周。每周测量大鼠收缩压(SBP)、舒张压(DBP)、平均动脉压(MAP);取血检测血清中甘油三酯(TG)、总胆固醇(TC)、尿酸(UA)、葡萄糖(GLU)、醛固酮(ALD)、肾素(Renin)、血管紧张素Ⅱ(Ang-Ⅱ)、血管紧张素转换酶(ACE)水平;ELISA法测定肾组织中Ang-Ⅱ及ACE水平;免疫组化法检测ACE、血管紧张素转换酶2(ACE2)蛋白表达;Western Blot法测定Ang-Ⅱ蛋白表达;HE染色观察心脏、胸主动脉及肾脏组织形态学改变。结果与模型组比,复方野菊花提取物能明显降低模型大鼠收缩压、舒张压及平均动脉压(P<0.05,P<0.01);降低血清TC、TG、UA及GLU含量(P<0.05,P<0.01);并能改善心脏、肾脏及胸主动脉组织形态学改变;可降低血清中ALD、Renin、Ang-Ⅱ、ACE水平(P<0.05,P<0.01);抑制肾脏中Ang-Ⅱ(P<0.01)、ACE(P<0.05,P<0.01)蛋白表达,增加ACE2蛋白表达。结论复方野菊花提取物具有改善代谢性高血压模型大鼠的作用,其降压机制可能与抑制过度激活的肾素-血管紧张素-醛固酮系统(RAAS)有关。 Objective To observe the effect and possible mechanism of compound Dendranthema indicum extract on metabolic hypertension model rats induced by high glucose,high-fat diet,and alcohol drinking.Methods The rats were fed with high glucose and high fat diet combined with alcohol gradient to prepare the metabolic hypertension model,and the compound Dendranthema indicum extract(5.25,3.50,1.75 g·kg^(-1))was given for 4 consecutive weeks after the model was successfully established.Systolic blood pressure(SBP),diastolic blood pressure(DBP)and mean arterial pressure(MAP)of rats were measured every week.Blood samples were taken to detect the levels of triglyceride(TG),total cholesterol(TC),uric acid(UA),glucose(GLU),aldosterone(ALD),renin,angiotensin 2(Ang-Ⅱ)and angiotensin converting enzyme(ACE).The levels of Ang-Ⅱand ACE in renal tissue were measured by ELISA,the protein expression of ACE and ACE2 was detected by immunohistochemistry,and the expression of Ang-Ⅱprotein was measured by Western Blot.HE staining was used to observe the histomorphological changes of heart,thoracic aorta and kidney.Results The results showed that compared with the model group,the compound Dendranthema indicum extract could significantly reduce SBP,DBP and MAP(P<0.05,P<0.01),reduce the contents of serum TC,TG,UA and GLU(P<0.05,P<0.01),and improve the histomorphological changes of heart,kidney and thoracic aorta.It can also reduce the levels of ALD,Renin,Ang-Ⅱand ACE in serum(P<0.05,P<0.01),inhibit Ang-Ⅱ(P<0.01)and ACE(P<0.05,P<0.01)protein expression in kidney and increase the expression of ACE2 protein.Conclusion In conclusion,compound Dendranthema indicum extract showed improvement in model rats with metabolic hypertension.Its antihypertensive mechanism may be related to the inhibition of overactivated RAAS.
