微小RNA-200c对人胰腺癌细胞株PANC-1中转化生长因子-β诱导基因-克隆3表达的影响 被引量:1

Effects of microRNA-200c on transforming growth factor-β-induced gene-human clone 3 expression in human pancreatic cancer strain PANC-1

摘要 目的观察微小RNA-200c(miR-200c)对人胰腺癌细胞株PANC-1中转化生长因子-β诱导基因-克隆3(βig-h3)表达的影响。方法将miR-200c前体片段和阴性对照片段分别转染到PANC-1细胞中。应用反转录-聚合酶链反应(RT-PCR)检测转染后的PANC-1细胞中miR-200c和βig-h3的表达。应用Transwell实验检测转染后PANC-1细胞的侵袭能力。采用t检验。结果转染miR-200c前体片段和阴性对照片段的PANC-1细胞中miR-200c的相对表达值分别为1.00±0.12和0.17±0.06,差异有统计学意义(t=3.008,P<0.05)。βig-h3的相对表达值分别为0.21±0.16和0.71±0.23,差异有统计学意义(t=2.236,P<0.05)。转染miR-200c前体片段和阴性对照片段的PANC-1细胞平均穿膜细胞数分别为(65.00±18.19)和(308.00±5.73)个,差异有统计学意义(t=2.107,P<0.05)。结论miR-200c可以抑制胰腺癌细胞的侵袭能力,可能与下调βig-h3的表达有关。 Objective To explore the effects of microRNA(miR)-200c on transforming growth factor(TGF)-β-induced gene-human clone 3(βig-h3)expression in human pancreatic cancer strain PANC-1.Methods MiR-200c precursor fragment and negative control fragment were transfected into human pancreatic cancer strain PANC-1.The reverse transcriptase-polymerase chain reaction(RT-PCR)was used to detect the expression of miR-200c andβig-h3 in PANC-1.Transwell invasion assay was conducted to detect the invasiveness.T test was used.Results The relative miR-200c expression level in PANC-1 of miR-200c precursor transfected group(1.00±0.12)was significantly higher than that of negative control group(0.17±0.06,t=3.008,P<0.05).The mRNA ofβig-h3 in miR-200c precursor transfected group was(0.21±0.16),and that in the negative control group was(0.71±0.23)with difference being significant between the two groups(t=2.236,P<0.05).The transmembrane number of miR-200c precursor transfected group[(65.00±18.19)cells]was les than that of negative control group[(308.00±5.73)cells,t=2.107,P<0.05].Conclusion Overexpression of miR-200c could inhibit invasiveness of PANC-1,which may be related with the down-regulation ofβig-h3 expression.
