摘要
本文采用三层高精度滴头,以明胶和海藻酸钠为胶皮,葵花籽油为载体,棕榈油为固态脂质保护层,制备含有副干酪乳杆菌的益生菌珠.考察了三层高精度滴头中固态脂质保护层在滴丸制备过程中对益生菌活性的保护作用,考察了胃肠消化过程中益生菌珠对胃肠道消化环境的耐受能力,评价了益生菌珠在4℃和室温下1个月储藏的储藏稳定性.结果表明,干燥后的三层高精度滴头制备的菌珠中活菌数为(8.97±0.01)lgCFU/g,菌体存活率为(87.48±2.41)%;经过胃部和小肠模拟消化后,菌珠的活菌数为(8.89±0.01)lgCFU/g和(8.68±0.04)lgCFU/g;在4℃及室温下储藏1年后,菌珠的菌活分别为(8.78±0.02)lgCFU/g和(8.69±0.02)lgCFU/g,存活率为(64.42±2.46)%和(52.60±2.42)%,显著高于双层滴头包埋菌珠.基于三层高精度滴头实现的液态油相、固态脂质及水凝胶结合的三层包埋菌珠,可以显著提高益生菌在加工、胃肠道消化及储藏过程中的活性,提供了一种有效的益生菌胃肠道递送方式.
In this paper,probiotic beads containing Lactobacillus paracasei were prepared by using three-layer high-precision drippers,gelatin and sodium alginate as rubber,sunflower seed oil as carrier and palm oil as solid lipid protective layer.The protective effect of solid lipid protective layer in three-layer high-precision dripper on probiotic activity in the preparation of dropping pills was investigated.The tolerance of probiotic beads to gastrointestinal digestive environment during gastrointestinal digestion was investigated.The storage stability of probiotic beads stored at 4℃and room temperature for one month was evaluated.The results showed that the number of viable bacteria in the beads prepared by the three-layer high-precision dripper after drying was(8.97±0.01)lgCFU/g,and the cell survival rate was(87.48±2.41)%.After simulated digestion of the stomach and small intestine,the viable counts of the beads were(8.89±0.01)lgCFU/g and(8.68±0.04)lgFU/g.After storage at 4℃and room temperature for 1 year,the bacterial activities of the beads were(8.78±0.02)lgCFU/g and(8.69±0.02)lgCFU/g,respectively.The survival rates were(64.42±2.46)%and(52.60±2.42)%respectively,which was significantly higher than that of the beads embedded in double-layer drippers.The three-layer embedded beads based on the combination of liquid oil phase,solid lipid and hydrogel can significantly improve the activity of probiotics in the process of processing,gastrointestinal digestion and storage,providing an effective way of probiotic gastrointestinal delivery.
