miR-124联合miR-570在宫颈癌不同病变阶段组织中的表达及其意义

Expression and significance of miR-124 and miR-570 in different pathological stages of cervical cancer

目的:探讨miR-124联合miR-570在宫颈癌早期诊断中的临床价值。方法:对2017年4月至2019年4月在潍坊市人民医院治疗的60例宫颈癌患者进行回顾性分析,将所取肿瘤标本作为实验组。选择同期体格检查确诊的57名健康女性的正常宫颈组织作为对照组。使用FIGO分期标准进行分期,逆转录-定量聚合酶链反应(qRT-PCR)检测miR-124和miR-570在组织中的表达,分析miR-124和miR-570单独的诊断价值及其在早期宫颈癌患者中的联合诊断价值。结果:实验组miR-124和miR-570水平低于对照组(P<0.05)。宫颈癌组织的不同阶段差异有统计学意义(P<0.05)。宫颈癌组织分期越晚,miR-124和miR-570的水平越低。miR-124和miR-570在早期宫颈癌的联合诊断中的灵敏度和曲线下面积(AUC)值显著高于单独的miR-124和miR-570。结论:miR-124联合miR-570检测较二者单独检测在宫颈癌早期诊断中更灵敏,对宫颈癌患者的治疗和预后判断有一定的应用潜能。

Objective: To investigate the clinical value of the combined detection of miR-124 and miR-570in the early diagnosis of cervical cancer. Methods: A retrospective analysis of 60 cervical cancer patients treated in the Weifang People’s Hospital in Shandong Province from April 2017 to April 2019was performed, and the lesion tissues were used as the experimental group. Normal cervical tissues from another 57 healthy females confirmed by physical examination at the same period were selected as the control group. The FIGO staging criteria were used for staging of the cervical cancer patients,reverse transcription-quantitative polymerase chain reaction(RT-qPCR) method was used for the detection of the expression of miR-124 and miR-570 in different tissues. The application value of miR-124 and miR-570 diagnosis alone and their combined diagnosis was evaluated in early cervical cancer patients. Results: The levels of miR-124 and miR-570 in the experimental group were lower than those in the control group(P<0. 05). The differences were significant in the different stages of cervical cancer tissues(P<0. 05). The later the staging of cervical cancer tissues were, the lower the levels of miR-124 and miR-570 were. The sensitivities and area under the ROC curve(AUC) values of miR-124 and miR-570 in the combined diagnosis of early cervical cancer were significantly higher than those of miR-124 and miR-570 alone. Conclusion: The detection of miR-124 combined with miR-570 is more sensitive than the detection of miR-124 and miR-570 alone in the early diagnosis of cervical cancer, and has certain application potential for the treatment development and prognosis judgement of cervical cancer patients.
