外周血CD226基因多态性与小儿流感病毒感染易感性的关联性 被引量:1

Relationship between CD226 gene polymorphism in peripheral blood and susceptibility to influenza virus infection in children

摘要 目的 探讨外周血CD226基因多态性与小儿流感病毒感染易感性的关系。方法 选择青岛市海慈医院儿科2018年1月-2021年1月收治的流感患儿73例,其中重症组29例和轻症组44例,同期医院健康体检儿童30名为对照组。收集入组儿童性别、年龄以及流感患儿病原类型、发病季节等临床资料。选择与流感相关的rs727088位点设计特异性引物,采用限制性片段长度多态性聚合酶链反应(PCR-RFLP)对目的片段进行扩增并分析其基因多态性分布。结果 重症组、轻症组和对照组CD226基因rs727088位点基因型分布均符合哈迪-温伯格遗传平衡(P>0.05);重症组、轻症组和对照组CD226基因rs727088位点GG基因型和A/G等位基因频率整体分布差异具有统计学意义(P<0.05),其中重症组和轻症组GG基因型频率(55.17%和47.73%)与G等位基因频率(63.79%和59.09%)均高于对照组(P<0.05);显性模型、隐性模型和加性模型遗传模式结果显示,重症组和轻症组不同基因型组合均差异无统计学意义。结论 CD226基因rs727088位点增加小儿流感病毒感染易感性,但对疾病严重程度无明显影响。 OBJECTIVE To explore the relationship between the CD226 gene polymorphism in peripheral blood and the susceptibility to influenza virus infection in children. METHODS A total of 73 children with influenza who were treated in department of pediatrics of Qingdao Hiser Hospital from Jan 2018 to Jan 2021 were enrolled in the study and divided into the severe group with 29 cases and the mild group with 44 cases, meanwhile, 30 children who received physical examination were set as the control group. The clinical data including gender, age, etiological types and onset seasons were collected from the children, the rs727088 locus that was associated with influenza was selected to design specific primers, the target fragments were amplified by polymerase-chain-reaction-restriction-fragment-length polymorphism(PCR-RFLP). RESULTS The distribution of genotypes at rs727088 locus of CD226 gene of the severe group, the mild group and the control group was in line with the Hardy-Weinberg genetic balance(P>0.05). There were significant differences in the frequencies of GG genotype and A/G allele at rs727088 locus of CD226 gene among the severe group, the mild group and the control group(P<0.05);the frequencies of GG genotype and G allele of the severe group and the mild group were respectively 55.17%, 47.73%, 63.79% and 59.09%, significantly higher than those of the control group(P<0.05). The results of genetic pattern analysis showed that under the dominant model, recessive model and additive model, there was no significant difference in the genotype combinations between the severe group and the mild group. CONCLUSION The rs727088 locus of CD226 gene may increase the susceptibility to influenza virus infection in the children but does not have significant effect on severity of disease.
