甘草悬浮细胞生产总黄酮条件优化及抗氧化性研究

Optimization of Production Conditions and Antioxidant Activity of Total Flavonoids From Glycyrrhiza uralensis Fisch.Suspension Cells

摘要 以甘草悬浮细胞为试验材料,采用响应面方法,研究了水解酪蛋白(CH)、2,4-D、6-BA对甘草悬浮细胞总黄酮得率的影响,以期获得甘草悬浮细胞生产总黄酮的最佳培养条件,为甘草悬浮细胞总黄酮的规模化生产提供参考依据。结果表明:当悬浮细胞培养基配方为MS液体培养基+CH 398.608 mg·L^(-1)+2,4-D 1.044 mg·L^(-1)+6-BA 1.325mg·L^(-1)时,模型预测的总黄酮最高得率为1.324%,验证试验的总黄酮得率为1.286%±0.03%,与预测值相差较小;甘草悬浮细胞总黄酮粗提液具有一定的抗氧化能力,且总抗氧化能力优于维生素C。 Taking Glycyrrhiza uralensis suspension cells as experimental material,the effects of hydrolyzed casein,2,4-D and 6-BA on the yield of total flavonoids from G.uralensis suspension cells were studied by response surface methodology,in order to obtain the optimal culture conditions for the production of total flavonoids from G.uralensis suspension cells,provide reference for production.The results showed that when the suspension cell culture medium was MS liquid medium+CH 398.608mg·L^(-1)+2,4-D 1.044mg·L^(-1)+6-BA 1.325mg·L^(-1),the maximum yield of total flavonoids predicted by the model was 1.324%,and the yield of total flavonoids in the validation test was 1.286%±0.03%,which was less different from the predicted value.The crude extract of total flavonoids from G.uralensis suspension cells had certain antioxidant capacity,and the total antioxidant capacity was better than vitamin C.
