饮片提取和配方颗粒调配的仲归颗粒中京尼平苷酸和阿魏酸含量差异研究

Study on the Difference of Contents of Geniposide acid and Ferulic Acid in Zhonggui Granules Prepared by Decoction and TCM Formula Granules

摘要 目的:使用全方饮片合煎液制备及配方颗粒调配仲归颗粒,通过比较两种样品中京尼平苷酸和阿魏酸的含量,研究各自制备方法的差异。方法:根据仲归颗粒组方,取全部药味加水煎煮,所得提取液制备颗粒剂;同时取相当于各药味生药量的中药配方颗粒,混匀制成混合颗粒剂;上述样品制备成供试品溶液,使用高效液相色谱法测定京尼平苷酸和阿魏酸的含量,使用色谱柱为Waters XDB C18(柱长250mm,直径4.6mm,粒径5μm),流动相使用乙腈(A)-0.1%磷酸(B),使用梯度洗脱,程序为:0~10min,10%-15%A;10~20min,15%-50%A;20~25min,50%-90%A;25~30min,90%-10%A;后运行10min;流速1.0mL/min,检测波长238nm(0~10min)、316nm(10~30min),柱温25℃,进样量10μL;共平行制备各10批样品进行测定。结果:10批饮片煎液制备的仲归颗粒中,阿魏酸的含量为0.4036~0.4987mg/g,京尼平苷酸的含量为0.1758~0.1942mg/g;10批配方颗粒调配的仲归颗粒中,上述两种成分的含量分别为0.1383~0.1894mg/g和0.3126~0.4650mg/g。结论:两种方法制备样品中目标成分的含量无明显差异,但饮片混合煎煮法所得颗粒含量相对较为接近,建议采用该方法制备仲归颗粒。 Objective:To study the differences of preparation methods by comparing the contents of geniposide acid and Ferulic Acid in Zhonggui granules prepared by decoction and TCM formula granules.Methods:According to the formulation of Zhonggui granules,all the ingredient were taken and decocted in water,and the obtained extract was used to prepare granules.At the same time,the TCM formula granules equivalent to the weigh of raw herbs were mixed and made into mixed granules.The samples were prepared and the contents of genipenic acid and Ferulic Acid were determined by HPLC on A Waters XDB C18 column(250 mm in length,4.6 mm in diameter,5μm in diameter).The mobile phase was acetonitrile(A)-0.1% phosphoric acid(B),followed by gradient elution.The procedure was 0~10 min,10%~15%A;10~20 min,15%~50%A;20~25 min,50%~90%A;25~30 min,90%~10%A;post run time was set at 10 min;The flow rate was 1.0 mL/min,the detection wavelength was 238 nm(0~10 min),316 nm(10~30 min),the column temperature was 25℃,and the sample size was 10μL.A total of 10 batches of samples were prepared in parallel for determination,respectively.Results:The contents of Ferulic Acid and geniposide acid were 0.4036~0.4987 mg/g and 0.1758~0.1942 mg/g in 10 batches of Zhonggui granules prepared by decoction.The contents of the above two components were 0.1383~0.1894 mg/g and0.3126~0.4650 mg/g in 10 batches of Zhonggui granules prepared by formula granules.Conclusion:There is no significant difference in the content of the target components in the samples prepared by the two methods,but the content of the particles obtained by the mixed decocting method is relatively close.It is suggested to prepare Zhonggui granules by this method.
