摘要
为了分析不同贮存方式和液体培养条件对越南油茶花粉萌率发及花粉管伸长的影响,以及解析越南油茶花粉萌发特性的个体差异,本研究以越南油茶花粉为材料,采用离体培养法研究了培养基组分、培养温度、贮藏方式及不同单株对花粉生长的影响。结果表明:越南油茶花粉在含100 g/L蔗糖和0.05 g/L硼酸的液体培养基中,25℃条件下培养,萌发效果最好;不同个体花粉萌率发及花粉管伸长差异极显著;越南油茶花粉贮存最佳方式为干燥后置于-20℃贮存。本研究结果可为解析越南油茶“花而不实”引起低产问题和研发人工授粉技术提供参考依据。
In order to analyze the effects of different storage methods and liquid culture conditions on pollen germination rate and pollen tube elongation of Camellia vietnamensis,and to analyze the individual differences in pollen germination characteristics of Camellia vietnamensis,this study uses the pollen of Camellia vietnamensis as the material,and uses the in vitro culture method to study the effects of medium components,culture temperature,storage methods and different individual plants on pollen growth.The results showed that the pollen of Camellia vietnamensis cultivated in liquid medium containing 100 g/L sucrose and 0.05 g/L boric acid at 25℃has the best germination effect.There are significant differences in pollen germination rate and pollen tube elongation among different individuals.The optimal storage method of pollen of Camellia vietnamensis is to store at-20℃after drying.The results of this study can provide a reference for analyzing the low yield problem caused by’flowering rather than fruiting’of Camellia vietnamensis and developing artificial pollination technology.
