摘要
中华鹅膏菌是一种具有独特气味且味道鲜美的野生食用菌。从福建武平县采集了一株中华鹅膏菌菌株WPAS01,通过ITS序列进行分子水平的种属鉴定,利用Illumina测序分析基因组特征,结合气相色谱质谱联用(GC-MS)和基因组序列解析其挥发性成分的主要成分及合成途径。结果表明,WPAS01菌株属于中华鹅膏菌的D分支,与源自福建漳平的HKAS100492菌株亲缘关系最近,单倍体基因组为47.54 Mb,杂合率为2.02%;从成熟子实体中检测出135种挥发性成分,其中15种在7种常见食用菌中也存在;其基因组中预测到所有莽草酸途径和艾氏通路合成苯乙醛的关键基因。
Amanita sinensis is a kind of delicious wild edible fungus with unique smell.The strain WPAS01 of A.sinensis was collected from Wuping county,Fujian province.The strain was used for species identification at molecular level by ITS sequence and for analyses of genomic characteristics based on Illumina sequencing,and the composition and synthetic pathway of its volatile compounds were analyzed by GAS chromatography-mass spectrometry(GC-MS)combined with the genome sequence.The results showed that strain WPAS01 is mostly closely related to strain HKAS100492 which collected from Zhangping,Fujian province,which both belonging to group D of A.sinensis.It is a heterokaryons with 2.02%heterozygosity rate and has a haploid genome of 47.54 Mb.Total 135 volatile components were detected from the mature fruit bodies,including 15 components also found in other common edible fungi.All key genes involved in Shikimic acid and Ehrlich pathway for biosynthesis of phenylacetaldehyde were predicted from genome of strain WPAS01.
