液相色谱-串联质谱法同时测定含乳饮料等食品中8种合成色素 被引量:6

Simultaneous Determination of 8 Synthetic Pigments in Dairy Beverages by Liquid Chromatography-tandem Mass Spectrometry

摘要 建立了超高效液相色谱-串联质谱法同时测定含乳饮料等食品中柠檬黄、日落黄、胭脂红、赤藓红、新红、诱惑红、亮蓝、酸性红的检测方法。样品采用氨化乙醇(乙醇∶氨水∶水=7∶2∶1(v/v/v))提取,纯水洗涤、聚酰胺粉吸附,氨化乙醇再次洗脱。洗脱液在70℃条件下减压旋蒸至干,用水溶解后调pH值至中性。经Waters ACQUITY UPLC BEH C18(2.1 mm×100 mm,1.7μm)分离。在流速0.3 mL/min下以乙腈-5 mmol/L乙酸铵溶液作为流动相梯度洗脱。实验采用三重四极杆电喷雾离子源(ESI源)电离-多反应监测(multiple reaction monitoring,MRM)-正离子监测模式检测。8种合成色素分别在20~500 mg/kg范围内线性关系良好,相关系数R2均大于0.990。加标回收率为79.6%~94.1%,RSD值为4.14%~14.32%。该方法提取率高,分析时间短,准确度高,检出限低,解决了复杂基质给液相色谱分离带来的干扰。 To establish an ultra performance liquid chromatography-tandem mass spectrometry method for simultaneous determination of lemon yellow,sunset yellow,carmine red,erythrum red,new red,lure red,bright blue and acid red in food containing milk drinks.The samples were extracted with ammoniated ethanol(ethanol∶ammonia∶water,volume ratio:7∶2∶1),washed with pure water,adsorbed with polyamide powder,eluted with ammoniated ethanol again.The eluent was evaporated to dry at 70℃,dissolved in water,and the pH was adjusted to neutral.Through ACQUITY UPLC BEH C18(2.1 mm×100 mm,1.7μm)separation,At the flow rate of 0.3 mL/min,acetonitrile-5 mmol/L ammonium acetate solution was used as mobile phase gradient elution.Triple quadrupole spray ion source(ESI source)ionization-multiple reaction monitoring(MRM)-positive ion monitoring mode was used in the experiment.The linear range of the 8 synthetic pigments was 20~500 mg/kg,and the correlation coefficients R2 were all greater than 0.990.The recoveries ranged from 79.6%to 94.1%,and the RSD ranged from 4.14%to 14.32%.This method has the advantages of high extraction rate,short analysis time,high accuracy and low detection limit,which solves the interference caused by complex matrix.
