摘要 CD47是一种在多种细胞上表达的多功能表面蛋白。作为膜受体参与许多正常和病理生理过程。CD47在急性髓细胞白血病细胞表面高表达,其与巨噬细胞上的信号调节蛋白α结合,介导“不要吃我”信号,抑制巨噬细胞的吞噬功能。通过CD47抑制剂阻断CD47/信号调节蛋白α信号通路的方法已成为肿瘤免疫治疗中的研究热点。本文就CD47诱导的免疫逃逸,靶向CD47的单克隆抗体在急性髓细胞白血病治疗中的研究进展进行综述。 CD47 is a multifunctional surface protein expressed in a wide range of cells.It acts as a membrane receptor involved in many normal and pathophysiological processes.CD47 is highly expressed on the surface of acute myeloid leukemia cells,binds to signal regulatory proteinα(SIRPα)on macrophages,mediates“don’t eat me”signal,and inhibits the phagocytic function of phagocytes.The method blocking the CD47/signal-regulatory proteinαsignaling pathway with CD47 inhibitors has become a hot topic in tumor immunotherapy.This article reviews the research advances of CD47 induced immune escape,monoclonal antibodies targeting CD47 in the treatment of acute myeloid leukemia.
