摘要
对新疆某地牛屠宰场采集的264份胴体拭子样品,选择特异性培养基并结合PCR法分离鉴定空肠弯曲菌,对空肠弯曲菌分离株测序后进行同源性分析,用结晶紫染色方法鉴定分离株空肠弯曲菌的生物膜形成情况。结果显示,共分离鉴定到空肠弯曲菌10株,总分离率为3.79%(10/264),牛屠宰场来源空肠弯曲菌同源性为94.0%~99.6%;10株空肠弯曲菌在结晶紫染色鉴定后,其OD 590nm值测定结果显示,72 h成膜能力显著高于48 h(P<0.001)和24 h(P<0.001),48 h成膜能力显著高于24 h(P<0.001)。表明该牛屠宰场空肠弯曲菌分离株亲缘性较近,存在一定程度水平传播污染,空肠弯曲菌分离株培养时间越长,其生物膜形成能力逐渐增强。
264 carcass swab samples collected from a cattle slaughterhouse in Xinjiang were used to isolate and identify Campylobacter jejuni by selecting specific medium and PCR.The homology of Campylobacter jejuni isolates was analyzed after sequencing,and the biofilm formation of Campylobacter jejuni isolates was identified by crystal violet staining.The results showed that 10 strains of Campylobacter jejuni were isolated and identified,and the total isolation rate was 3.79%(10/264).The homology of Campylobacter jejuni from cattle slaughterhouse was 94.0%-99.6%;After 10 strains of Campylobacter jejuni were identified by crystal violet staining,the OD_(590nm) value showed that the film-forming ability at 72 h was significantly higher than that at 48 h(P<0.001)and 24 h(P<0.001),and the film-forming ability at 48 h was significantly higher than that at 24 h(P<0.001).This study showed that the Campylobacter jejuni isolates from the cattle slaughterhouse were closely related,and there was a certain degree of horizontal transmission pollution of Campylobacter jejuni.The longer the culture time of Campylobacter jejuni isolates,its biofilm forming ability gradually increased.
