摘要
目的探究沉香线香熏香助睡眠作用及其作用机制。方法实验动物随机分为空白对照组、氯苯丙氨酸模型组、地西泮对照组(2.5 mg·kg^(-1))及熏香吸入低剂量组(0.25 g·kg^(-1))、中剂量组(0.5 g·kg^(-1))、高剂量组(1.0 g·kg^(-1));采用自制熏香仪熏点吸入的给药方法,1次/天,1 h/次,连续7天。观察沉香线香熏点吸入后对睡眠的影响;观察小鼠行为学变化并进行评价;采用酶联免疫吸附测定法(ELISA)测定小鼠脑内伽马氨基丁酸(GABAA)、谷氨酸(Glu)、五羟色胺(5-HT)、腺苷(AD)等递质及对GABAA/Glu值的影响;采用免疫印迹法(Western Blot)检测对谷氨酸能系统代谢和转运相关蛋白——谷氨酸受体(GluR1)及囊泡谷氨酸转运蛋白(VGluT1)表达的影响。结果在一定剂量下,沉香线香熏香吸入可显著提高入睡效率和时长(P<0.05),同时减少其自主活动时间(P<0.05);熏香组可显著升高5-HT、Glu、GABAA及腺苷分泌水平(P<0.01或P<0.05),同时上调GABAA/Glu的比值(P<0.01或P<0.05);沉香线香熏香还可显著升高海马组织中GluR1及VGluT1相关蛋白的表达。结论沉香线香熏香吸入可显著抑制失眠小鼠的自主活动,较好改善睡眠状况,作用机制可能与调节脑内5-HT递质含量、调控GABA-Glu分泌平衡及Glu合成代谢和转运有关。
Objective To explore the sleep-enhancing effect and its mechanism of agarwood incense inhalation.Methods The experiment included blank group,Chlorophenylalanine group,Diazepam group,agarwood incense low group(0.25 g·kg^(-1)),agarwood incense middle group(0.5 g·kg^(-1))and agarwood incense high group(1.0 g·kg^(-1)),positive group was injected diazepam(10 ml/kg).Agarwood incense group was inhaled in the incense cable once an hour.Respective drugs were given except the blank and model group.Then sleep aid experiment was employed to assess the sleep quality,and the autonomic activities of rats in each group was tested;We probed the index as 5-HT,Glu,GABAA,AD and GABAA/Glu of rats,and explored the protein expressions of Gln and GluT1 in the hippocampus.Results The agarwood incense could significantly get the rats to sleep(P<0.05)and the same time extend the time(P<0.05);It decreased the rat autonomic activities,including reduced total distance and average velocity(P<0.05),and increased rest time(P<0.001);Inhaling the agarwood incense could increase the level of 5-HT,Glu,GABA and AD(P<0.01 or P<0.05);Meanwhile,GABAA/Glu(P<0.01 or P<0.05),and the protein expression of GluR1 and VGluT1 was increased.Conclusion Inhaling the agarwood incense can decrease the general activity of rats and significantly improving-sleep,and the mechanism of action may be related to adjusting the balance of GABAA/Glu and synthetic metabolism in Glu system pathway.
