基于PLC-DAG-PKC信号通路探讨背俞指针疗法对胃食管反流病的影响 被引量:7

Effect of Back-Shu Finger Therapeutics on Gastroesophageal Reflux Disease Based on PLC-DAG-PKC Signal Pathway

摘要 目的观察背俞指针疗法对胃食管反流病(Gastroesophageal reflux disease,GERD)大鼠PLCDAG-PKC信号通路的影响。方法将50只大鼠随机分为空白组、模型组、指针组、西药组,采用食管支架置入术构建GERD大鼠模型,西药组予以兰索拉唑肠溶片联合枸橼酸莫沙必利分散片灌胃,指针组施以背俞指针疗法进行干预,疗程均为2周。干预期间,观察各组大鼠的一般状态。干预结束后,光镜下观察大鼠食管下段粘膜病理组织学改变,免疫组化法(Immunohistochemistry,IHC)和免疫印迹法(Western Blot,WB)观察胃运动起搏区平滑肌组织PLC、DAG、PKC蛋白表达。结果模型组大鼠一般状态较差,口角处可见反流少量食物残渣,光镜下可见食管鳞状上皮基底细胞层增厚,固有层乳头延长伴炎性细胞浸润,细胞间隙增宽。IHC和WB结果示模型组PLC、DAG蛋白表达均较空白组降低(P<0.05),PKC蛋白表达较空白组呈降低趋势但无统计学意义(P>0.05);西药组和指针组病理学观察可见食管粘膜结构基本恢复正常,鳞状上皮修复完整,基底细胞呈单层排列,未见明显炎症细胞浸润,IHC结果示西药组和指针组胃运动起搏区平滑肌组织PLC、DAG、PKC蛋白分布较模型组升高,差异有统计学意义(P<0.01)。结论背俞指针疗法能有效修复GERD大鼠食管粘膜结构,其机制可能与调控PLC-DAG-PKC信号通路相关。 Objective To observe the effect of Back-Shu Finger Therapeutics on PLC-DAG-PKC signal pathway in rats with gastroesophageal reflux disease(GERD).Methods 50 rats were randomly divided into the blank group, the model group, the finger acupuncture group and the western medicine group. The GERD rat model was established by esophageal stent implantation. The western medicine group was given lansoprazole enteric-coated tablets combined with mosapride citrate dispersible tablets by gavage. The pointer group was intervened by back-shu finger acupuncture therapy, with a course of two weeks. During the intervention, the general state of rats in each group was observed. After the intervention, the histopathological changes of the lower esophageal mucosa were observed under light microscope,and the expressions of PLC, DAG and PKC in the gastric pacing area were observed by immunohistochemistry(IHC) and Western Blot(WB).Results The rats in the model group were generally in poor condition. A small amount of reflux food residue could be seen at the mouth angle. Under the light microscope, the basal cell layer of esophageal squamous epithelium was thickened. The papilla of lamina propria was prolonged with inflammatory cell infiltration, and the intercellular space was widened. IHC and WB results showed that the expression of PLC and DAG protein in the model group was lower than that in the blank group(P<0.05), and the expression of PKC protein showed a decreasing trend but no statistical significance(P>0.05). The pathological observation of western medicine group and pointer group showed that the structure of esophageal mucosa was basically restored to normal, and the squamous epithelium was repaired completely. The basal cells were arranged in monolayer, and there was no obvious inflammatory cell infiltration. IHC results showed that the distribution of PLC, DAG and PKC proteins in the western medicine group and the finger acupuncture group was higher than that in the model group, and the difference was statistically significant(P<0.01).Conclusion Back-Shu Finger Therapeutics can effectively repair the esophageal mucosal structure of GERD rats, and its mechanism may be related to the regulation of PLC-DAG-PKC signaling pathway.
