微生物发酵联用折光仪检测糖蜜含糖量的研究 被引量:1

摘要 目的:建立一种准确、高效的糖蜜含糖量检测方法,弥补该领域的空白,规范糖蜜市场,为以糖蜜为原料的发酵生产企业提供低成本检测糖蜜含糖量的检验方法和思路。方法:同时使用新构建的微生物发酵联用折光仪和多糖检测试剂盒法检测,比较两者偏差值;采用葡萄糖加标回收、非糖可溶性固形物干粉VINASE添加考察新建方法的准确性和可靠性。结果:以酿酒酵母、嗜渗酵母为发酵菌株实验组,以嗜渗酵母为发酵菌株实验结果更优,偏差范围在-1.50%~2.00%,平均绝对偏差为0.46%;葡萄糖加标回收实验,0.5 g实验组回收率在84.0%~106.0%,平均回收率高达90.8%;2.0 g实验组回收率在94.8%~105.2%,平均回收率高达100.5%;VINASE实验组,0.5 g实验组偏差在-2.4%~1.1%,平均绝对偏差仅为0.4%;2.0 g实验组偏差在-3.5%~1.7%,平均绝对偏差仅为-1.1%;对比多糖检测试剂盒法,微生物发酵联用折光仪法对核心设备及试剂几乎无特别要求,不依赖于进口试剂。结论:新建立的微生物发酵联用折光仪法相比传统多糖检测试剂盒法,是一种成本极低、效率较高的糖蜜含糖量检测方法,具有优异的准确性、稳定性,检测门槛低,可被广泛应用于糖蜜生产、交易、加工行业。 Objective:To establish an accurate and efficient molasses sugar content detection method,to fill the gaps in this field,to standardize the molasses market,and to provide testing methods and ideas that can detect the sugar content of molasses at low cost for fermentation production enterprises using molasses as raw materials.Method:The newly constructed microbial fermentation combined refractometer and polysaccharide kit were used to detect the deviation between the two methods.The accuracy and reliability of the new method were evaluated by glucose standard recovery and non-sugar soluble solid dry VINASE addition.Result:Saccharomyces cerevisiae and lyophilic yeast were used as fermentation strains in the experimental group,and lyophilic yeast was used as fermentation strain.The experimental results were better,with the deviation ranging from-1.50%~2.00%,and the average absolute deviation was 0.46%.In the glucose spiked recovery experiment,the recovery rate of 0.5g group was 84.0%~106.0%,and the average recovery rate was up to 90.8%.The recoveries of 2.0 g group were 94.8%~105.2%,and the average recoveries were 100.5%.In VINASE group,the deviation of 0.5 g group was between-2.4%~1.1%,and the average absolute deviation was only 0.4%.The deviation of 2.0 g experimental group was between-3.5%~1.7%,and the average absolute deviation was only-1.1%.Compared with polysaccharide detection kit method, microbial fermentation combined refractometer method has almost no special requirements for core equipment and reagents, and does not rely on imported reagents. Conclusion: Compared with traditional polysaccharide detection kit method, the newly established microbial fermentation combined refractometer method is a very low test cost and high-efficiency method for the detection of sugar content in molasses, with excellent accuracy, stability and low detection threshold, can be widely used in molasses production, trading and processing industries.
