NOD蛋白在浆细胞性乳腺炎患者病变组织中的表达

摘要 目的探讨核苷酸寡聚化结构域受体(NOD)1蛋白、NOD2蛋白与浆细胞性乳腺炎(plasma cell mastitis,PCM)的相关性。方法收集银川市妇幼保健院2018年1月至2020年1月入院的26例PCM住院患者的病变组织,采集PCM患者炎性肿块组织和距离炎性肿块边缘≥3 cm且镜检为正常乳腺组织的样本。通过实时荧光定量聚合酶链式反应(qRT-PCR)和蛋白质印迹(Western blot)法检测PCM组织和正常乳腺组织中NOD1、NOD2 mRNA和蛋白的表达,并分析两者的相关性。结果qRT-PCR检测PCM患者炎性肿块中NOD1、NOD2 mRNA水平均较正常组织高(P均<0.05),且NOD1与NOD2 mRNA表达呈正相关(r=0.9632,P<0.05)。Western blot检测结果显示与正常组织相比,PCM患者炎性肿块中NOD1和NOD2蛋白表达水平均升高(P均<0.05);在PCM患者炎性肿块中NOD1与NOD2蛋白表达水平呈正相关(r=0.9972,P<0.05)。结论PCM组织中NOD1和NOD2蛋白高表达,可能与PCM发病有关。 Objective To investigate the relationship between NOD1 protein,NOD2 proteins and plasma cell mastitis(PCM).Methods The pathological tissues of 26 hospitalized patients with PCM in Yinchuan Maternal and Child Health Hospital from January 2018 to January 2020 were collected.The samples of inflammatory mass tissue and normal breast tissue from PCM patients≥3 cm away from the edge of inflammatory mass were collected.The expressions of NOD1 and NOD2 mRNA and protein in PCM and normal breast tissues were detected by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR)and Western blot,and the correlation between them was analyzed.Results The levels of NOD1 and NOD2 mRNA in inflammatory masses detected by qRT-PCR were higher than those in normal tissues(P all<0.05),and there was a positive correlation between NOD1 and NOD2 mRNA(r=0.9632,P<0.05).Western blot results displayed that compared with normal tissues,the expression levels of NOD1 and NOD2 proteins in PCM patients with inflammatory masses were increased(P all<0.05);NOD1 and NOD2 protein expression levels were positively correlated in PCM inflammatory masses(r=0.9972,P<0.05).Conclusion The significant overexpression of NOD1 and NOD2 proteins in PCM may be related to the pathogenesis.
