摘要
目的:探讨癌症相关区域长链非编码RNA5(CARLo-5)在食管癌组织中的表达及临床意义。方法:选取2015年3月至2016年10月接受食管癌根治术的58例患者,每例均取癌组织及癌旁组织(距癌组织>5cm),采用实时荧光定量PCR(Realtime-PCR)法检测患者癌组织及对应癌旁组织标本CARLo-5表达水平,比较CARLo-5在食管癌组织和食管癌旁上皮组织中的相对表达量,并分析CARLo-5表达与食管癌患者临床特征及预后的关系。结果:食管癌组织中CARLo-5相对表达量(4.25±9.02)明显高于配对癌旁组织(1.09±0.95),差异有统计学意义(t=2.653,P<0.05)。食管癌组织中CARLo-5表达与患者年龄、性别、肿瘤分化程度、位置和大小无关(P>0.05),与肿瘤TNM分期、淋巴结转移情况有关(P<0.05)。术后随访显示58例患者中死亡39例,占67.24%。CARLo-5高表达组(34例,生存6例)生存率显著低于CARLo-5低表达组(24例,生存13例)(χ^(2)=6.941,P<0.05);高表达组平均生存期为(24.39±1.68)月,低表达组平均生存期为(31.42±1.48)月,高表达组生存期显著短于低表达组(t=2.653,P<0.05)。单因素分析显示TNM分期、淋巴结转移、CARLo-5为影响EC患者OS的危险因素(P<0.05),COX多因素回归分析显示淋巴结转移、CARLo-5表达水平是影响OS的独立危险因素(P<0.05)。结论:该研究提示食管癌组织中CARLo-5表达水平显著上调,且与患者肿瘤TNM分期、淋巴结转移及预后密切相关;CARLo-5高表达可能参与食管癌发生发展,可用于预测食管癌患者预后。
Objective:To investigate the expression and clinical significance of long-chain non-coding RNA5(CARLo-5)in esophageal cancer tissues.Methods:Selecting 58 patients who underwent radical esophagectomy from March 2015 to October 2016.In each case,cancer tissues and adjacent tissues(>5 cm away from the cancer tissues)were taken and Real-time Quantitative polymerase chain reaction(Realtime-PCR)was used to detect the expression level of CARLo-5 in cancer tissues and corresponding adjacent tissues.Comparing the relative expression levels of CARLo-5 in cancer tissues and normal esophageal epithelial tissues,and analyzing the expression of CARLo 5 and the relationship among clinical features and prognosis of esophageal cancer patients.Results:The relative expression level of CARLo-5 in esophageal cancer tissues was up-regulated(4.25±9.02),which was significantly higher than that in corresponding adjacent tissues(1.09±0.95).The expression of CARLo-5 in esophageal cancer tissues was independent of age,gender,tumor differentiation,location and size(P>0.05),and was related to TNM stage and lymph node metastasis(P<0.05).Postoperative follow-up showed that 39 of the 58 patients died,accounting for 67.24%.The survival rate of the group with high expression of CARLo-5(34 cases,6 survival cases)was significantly lower than that of the group with low expression of CARLo-5(24 cases,13 survival cases)(χ^(2)=6.941,P<0.05).The average survival time of the high-expression group was(24.39±1.68)months,and that of the low-expression group was(31.42±1.48)months.The survival time of the high-expression group was significantly shorter than that of the low-expression group(t=2.653,P<0.05).Univariate analysis showed that TNM stage,lymph node metastasis and CARLo-5 were risk factors affecting OS in EC patients(P<0.05).COX multivariate regression analysis showed that lymph node metastasis and CARLo-5 expression level were independent risk factors affecting OS(P<0.05).Conclusion:This study suggests that the expression level of CARLo-5 in esophageal cancer tissues is significantly up-regulated,and is closely related to the TNM stage,lymph node metastasis and prognosis of the patients.High expression of CARLo-5 may be involved in the development of esophageal cancer and can be used to predict the prognosis of patients with esophageal cancer.
