摘要
研究宽筋藤的化学成分。宽筋藤干燥药材用70%乙醇回流提取,所得浸膏依次用氯仿、乙酸乙酯和正丁醇萃取。采用硅胶、ODS、Sephadex LH-20、大孔树脂等方法对宽筋藤的氯仿部位和正丁醇部位进行分离纯化,通过核磁共振波谱、质谱等现代波谱技术对分离得到的化合物进行结构鉴定。结果从宽筋藤的70%乙醇提取物中分离得到13个化合物,其中从氯仿部位分离得到4个化合物,分别为2-(2′-羟基二十四碳酰氨基)十八碳-1,3,4-三醇(1)、1,2-di-O-palmitoyl-3-O-β-D-galactopyranosyl glycerol(2)、soyacerebrosidesⅠ和Ⅱ(3)、tinocordiside(4);从正丁醇部位分离得到9个化合物,分别为反式丁香苷(5)、3,4-二甲氧基苯基-β-D-葡萄糖苷(6)、tinosinen(7)、5-羟甲基-2-呋喃丙烯酸(8)、tinosineside A(9)、tinosposide A(10)、异落叶松脂素-9-O-β-D-葡萄糖苷(11)、开环异落叶松脂素-9-O-β-D-葡萄糖苷(12)、L-焦谷氨酸(13)。其中,化合物8作为天然产物首次报道,化合物1、2、3、6、11为首次从防己科植物中分离得到,化合物13为首次从宽筋藤中分离得到。此外,对分离得到的13个化合物进行了抑制脂多糖诱导RAW 264.7细胞释放NO活性的测定。
Study on the chemical constituents from Tinosporae sinenisis(Lour.)Merr.The shattered stem of T.sinenisis was extracted with 70%ethanol under reflux.The crude extract was extracted with chloroform,ethyl acetate and n-butanol,successively.Thirteen compounds were isolated and purified by silica gel,ODS,Sephadex LH-20 and macroporous resin from chloroform and n-butanol fractions of T.sinenisis.Their structures were elucidated via the interpretation of nuclear magnetic spectrometry,mass spectrometry and other spectral analyses.Thirteen compounds were identified as 2-(2′-hydroxytetracosanoylamino)octadecane-1,3,4-triol(1),1,2-di-O-palmitoyl-3-O-β-D-galactopyranosyl glycerol(2),soyacerebrosidesⅠandⅡ(3),tinocordiside(4),trans-syringin(5),3,4-dimethoxyphenyl-β-D-glucoside(6),tinosinen(7),5-hydroxymethyl-2-furanacrylic acid(8),tinosineside A(9),tinosposide A(10),isolariciresinol-9-O-β-D-glucopyranoside(11),secoisolariciresinol-9-O-β-D-glucopyranoside(12)and L-pyroglutamic acid(13).Compounds 1-4 were isoalted from chloroform fraction,and compounds 5-13 were isolated from n-butanol fraction.Compound 8 is the first report from natural sources,compounds 1,2,3,6,11 are newly isolated from the family Menispermaceae,compound 13 is newly isolated from T.sinenisis.In addition,all the 13 compounds were evaluated for their inhibitory effects in lipopolysaccharide-induced nitric oxide production in RAW 264.7 cells.
