维生素D对小鼠结核感染后获得性免疫的影响

摘要 目的探讨维生素D对小鼠结核感染获得性免疫的影响及机制.方法将BALB/c小鼠分为对照组(n=20)、感染组(n=20)、维生素D干预组(n=20),分析各组小鼠外周血CD3^(+)T、CD4^(+)T和CD8^(+)T细胞比例及CD4^(+)/CD8^(+)T细胞比例,Elisa检测IFN-γ、IL-4、IL-17水平以及TGF-β.结果与对照组比较,感染组和维生素D干预组的CD3^(+)T、CD4^(+)T细胞比例增加(P<0.01),CD8^(+)T细胞降低(P<0.01),CD4^(+)/CD8^(+)T细胞比例升高(P<0.01).ELISA结果表明,维生素D干预组和感染组小鼠IFN-γ、IL-17、TGF-β升高,且维生素D干预组升高更明显,差异有统计学意义(P<0.001).结论维生素D可能对小鼠结核感染后免疫细胞平衡产生影响并介导免疫细胞因子释放. Objective To investigate the effect and mechanism of vitamin D on acquired immunity of tuberculosis infection in mice.Methods BALB/C mice were divided into control group(n=20),infection group(n=20),and vitamin D intervention infection group(n=20).The proportion of CD3^(+)T,CD4^(+)T and CD8^(+)T cells in peripheral blood and the proportion of CD4^(+)/CD8^(+)T cells in each group were analyzed.IFN-γ,IL-4,IL-17 and TGF-βwere detected by Elisa.Results Compared with the control group,the proportion of CD3^(+)T and CD4^(+)T cells was significantly increased in the infection group and the vitamin D intervention group(P<0.01),CD8^(+)T cells were significantly decreased(P<0.01),and CD4^(+)/CD8^(+)T cell ratio increased significantly(P<0.01).ELISA results showed that IFN-γ,IL-17 and TGF-βwere increased in both vitamin D intervention group and infection group,but significantly increased in vitamin D intervention group with statistical significance(P<0.001).Conclusion Vitamin D may affect the balance of immune cells and mediate the release of immune cytokines after tuberculosis infection in mice.
