白花蛇舌草-半枝莲药对乙酸乙酯组分对胃癌SGC-7901细胞周期的影响 被引量:10

摘要 目的:探讨白花蛇舌草-半枝莲药对乙酸乙酯组分对胃癌SGC-7901细胞周期的影响。方法:将胃癌SGC-7901细胞分为对照组(0 mg·L^(-1))及白花蛇舌草-半枝莲药对乙酸乙酯组分低(10 mg·L^(-1))、中(30 mg·L^(-1))、高(50 mg·L^(-1))剂量组,分别培养12 h、24 h、48 h后采用CCK-8法检测细胞增殖能力;平板细胞克隆形成实验检测SGC-7901细胞克隆形成能力;流式细胞术检测细胞周期变化;Western Blot检测细胞中细胞周期蛋白D1(Cyclin D1)及c-Myc蛋白表达水平。结果:与对照组比较,白花蛇舌草-半枝莲药对乙酸乙酯组分各剂量组细胞增殖抑制率显著升高(P<0.05),细胞集落形成率显著降低(P<0.05),G0/G1期细胞比例显著升高(P<0.05),G2/M期细胞比例显著降低(P<0.05),Cyclin D1及c-Myc蛋白表达水平显著降低(P<0.05),且呈剂量依赖性。结论:白花蛇舌草-半枝莲药对乙酸乙酯组分可将细胞阻滞于G0/G1期进而抑制SGC-7901细胞增殖,其作用与下调Cyclin D1、c-Myc蛋白表达水平有关。 Objective:To investigate the effect of ethyl acetate components of Baihuasheshecao(Hedyotis diffusa-Scutellaria barbata)on the cell cycle of gastric cancer SGC-7901.Methods:Gastric cancer SGC-7901 cells were divided into control group(0 mg·L^(-1)),ethyl acetate components of hedyotis diffusa-Scutellaria barbata with low(10 mg·L^(-1)),medium(30 mg·L^(-1))and high(50 mg·L^(-1))dose groups.After culturing for 12 h,24 h,and 48 h,the cell proliferation ability was detected by the CCK-8 method.The colony formation ability of SGC-7901 cells was detected by plate cell colony formation assay.Cell cycle changes were detected by flow cytometry.And Western Blot was used to detect the protein expression levels of Cyclin D1 and cellular myelocytomatosis oncogene(c-Myc)in cells.Results:Compared with the control group,the inhibition rate of ethyl acetate components of hedyotis diffusa-Scutellaria barbata on the cell proliferation of each dose group was significantly increased(P<0.05),and the colony formation rate of cells was significantly decreased(P<0.05),the proportion of cells in G0/G1 phase was significantly increased(P<0.05),and the proportion of cells in G2/M phase was significantly decreased(P<0.05).Cyclin D1 and c-Myc protein expression levels Significantly decreased(P<0.05)in a dose-dependent manner.Conclusion:Ethyl acetate components of hedyotis diffusa-Scutellaria barbata can inhibit the proliferation of SGC-7901 cells by blocking the cells in G0/G1 phase,and its effect is related to the down-regulation of Cyclin D1 and c-Myc protein expression levels.
