摘要
目的 优选莲子超微粉的制备工艺。方法 以粉碎时间、烘干时间、初始粒度为考察因素,以莲子超微粉累计粒度分布百分数达到50%对应的粒径(D)为评价指标,采用Box-Behnken响应面法优选最佳制备工艺,并将最佳工艺制得的莲子超微粉进行薄层鉴别及水分、总灰分测定。结果 优选的最佳制备工艺为原药材粉碎3 min,过5号筛,60℃烘干1 h。优选工艺制备的3批莲子超微粉的平均D为(710.33±2.87)nm,与预测值706.67 nm接近。所制备的莲子超微粉的水分为5.41%~8.10%,总灰分为2.67%~4.76%,薄层色谱图显色清晰、分离度好、专属性强。结论 Box-Behnken响应面法优选的莲子超微粉制备工艺合理、可行。
Objective To optimize the preparation process of Nelumbinis Semen Ultrafine Powder.MethodsThe Box-Behnken design-response surface methodology (BBD-RSM) was used to optimize the preparati process of Nelumbinis Semen Ultrafine Powder with the grinding time,drying time and initial granule size as the investigation factors,and with the granule size at 50%of cumulative granule size distribution (D) of Nelumbinis Semen Ultrafine Powder as the evaluation index.The Nelumbinis Semen Ultrafine Powder prepared by the optimal process was identified by the thin-layer chromatography (TLC) method,and the contents of moisture and total ash of Nelumbinis Semen Ultrafine Powder were determined.ResultsThe optimal preparation process was as follows:the raw materials were ground for 3 min,passed through No.5 sieve,and dried at 60℃for 1 h.The average Dof the three batches of Nelumbinis Semen Ultrafine Powder prepared by the optimal process was (710.33±2.87)nm,which was close to the predicted Dof 706.67 nm.The moisture content of prepared Nelumbinis Semen Ultrafine Powder was in the range of 5.41%-8.10%,and the total ash content of prepared Nelumbinis Semen Ultrafine Powder was in the range of 2.67%-4.76%.The TLC chromatograms showed clear color,good separation and strong specificity.ConclusionThe preparation process of Nelumbinis Semen Ultrafine Powder optimized by the BBD-RSM is reasonable and feasible.
