补肾清热育阴汤对非肥胖型糖尿病小鼠唾液分泌量及颌下腺IL-1β、IL-22含量的影响

Effects of Bushen Qingre Yuyin Decoction on saliva secretion and IL-1β and IL-22 in submandibular gland of NOD mice

摘要 目的:观察补肾清热育阴汤对非肥胖型糖尿病(NOD)小鼠唾液分泌量及颌下腺白细胞介素(IL)-1β、IL-22含量的影响。方法:将50只8周龄NOD小鼠随机分为模型组,羟氯喹组,中药低、中、高剂量组,每组10只。另设10只同周龄BALB/c小鼠作为正常组。中药低、中、高剂量组分别灌胃补肾育阴清热汤20、40、80 g/kg,羟氯喹组给予羟氯喹溶液80 mg/kg灌胃,正常组与模型组给予等量的0.9%氯化钠溶液,每日1次,持续给药12周。每2周评估1次小鼠唾液分泌量,采用ELISA法检测小鼠颌下腺IL-1β的含量,采用免疫组化法检测小鼠颌下腺IL-22水平。结果:实验第6周后,各给药组的唾液分泌量均显著多于模型组(P<0.05);实验第12周,与羟氯喹组比较,中药中、高剂量组的唾液分泌量显著增多(P<0.05)。与模型组比较,中药高剂量组小鼠颌下腺IL-1β含量显著降低(P<0.05),各给药组小鼠颌下腺IL-22含量显著降低(P<0.01)。结论:补肾清热育阴汤能够有效增加唾液分泌量,并且能下调颌下腺IL-1β、IL-22含量。 Objective:To observe the effects of Bushen Qingre Yuyin Decoction on salivary secretion and submandibular gland IL-1β and IL-22 contents in NOD mice.Methods:Fifty 8-week-old NOD mice were randomly divided into model group,hydroxychloroquine group,and low-dose,medium-dose and high-dose Chinese medicine groups,with 10 mice in each group.Another 10 BALB/c mice of the same age were set as the normal group.The low-dose,medium-dose and high-dose Chinese medicine groups were given 20,40,and 80 g/kg of Bushen Yuyin Qingre Decoction,respectively,the hydroxychloroquine group was given 80 mg/kg of hydroxychloroquine solution,and the normal group and the model group were given the same amount of 0.9% sodium chloride solution once daily for 12 weeks.The saliva secretion of mice was evaluated every 2 weeks.The content of IL-1β in the submandibular gland of mice was detected by ELISA,and the level of IL-22 in the submandibular gland of mice was detected by immunohistochemistry.Results:After the 6th week of the experiment,the saliva secretion of each administration group was significantly higher than that of the model group(P<0.05).In the 12th week of the experiment,compared with the hydroxychloroquine group,the saliva secretion of the medium and high dose Chinese medicine groups increased significantly(P<0.05).Compared with the model group,the content of IL-1β in the submandibular gland of the mice in the high-dose Chinese medicine group was significantly decreased(P<0.05),and the content of IL-22 in the submandibular gland of the mice in each administration group was significantly decreased(P<0.01).Conclusion:Bushen Qingre Yuyin Decoction can effectively increase salivary secretion and down-regulate the contents of IL-1β and IL-22 in submandibular gland.
