健脾解毒汤对脾虚湿盛型寻常型银屑病小鼠白细胞介素-17及其相关炎症因子的影响 被引量:4

Effects of Jianpi Jiedu Decoction on interleukin-17 in mice with psoriasis vulgaris of spleen deficiency and dampness excess type and its related inflammation factors

摘要 目的:观察健脾解毒汤及拆方对脾虚湿盛型寻常型银屑病小鼠白细胞介素-17(IL-17)及其相关炎症因子[γ-干扰素(IFN-γ)、白细胞介素-6(IL-6)]的影响。方法:70只SPF级雄性ICR小鼠随机分为空白对照组、脾虚模型组、脾虚+银屑病组、阿维A胶囊组、解毒组、健脾组和健脾解毒汤组,每组10只。给予大黄水煎液灌胃制作脾虚湿盛动物模型,5%咪喹莫特软膏外用诱导银屑病,造模完成后采集静脉血并取皮损组织。ELISA法检测小鼠血清中IL-17、IFN-γ、IL-6表达水平;HE染色观察皮损组织病理变化。结果:阿维A胶囊组、解毒汤组、健脾汤组和健脾解毒汤组之间有明显差异性(P<0.05),健脾解毒汤组效果最好。各药物干预组IFN-γ、IL-6、IL-17的表达量与空白对照组比较,均显著升高(P<0.05),而与脾虚模型组和脾虚+银屑病组比较,均显著降低(P<0.05)。脾虚+银屑病组出现明显银屑病样皮损状态,各治疗组有不同程度的缓解,其中健脾解毒汤组优于其他各治疗组。结论:健脾解毒汤可以调节IFN-γ、IL-6、IL-17的表达水平,可能是其治疗银屑病新的作用靶点。 Objective:To observe the effect of Jianpi Jiedu Decoction and its disassembled prescriptions on interleukin-17(IL-17) in mice with psoriasis vulgaris of spleen deficiency and dampness excess type symptomatic and its related inflammation factors as interferon-γ(IFN-γ) and interleukin-6(IL-6).Methods:A total of 70 male ICR mice of SPF grade were randomly divided into blank control group,spleen deficiency model group,spleen deficiency psoriasis group,Acitretin Capsule group,Jiedu group,Jianpi group and Jianpi Jiedu Decoction group,with 10 rats in each group.The animal model of spleen deficiency and dampness was made by intragastric administration of liquid.Psoriasis was induced by external application of 5% imiquimod ointment.After the completion of modeling,venous blood was collected and skin lesions were taken.ELISA determination of IL-17,IFN-γ,IL-6 expression level in mouse serum;HE staining was used to observe the histopathological changes of skin lesions.Results:Acitretin Capsule group,Jiedu group,Jianpi group and Jianpi Jiedu Decoction group,There was significant difference among the four groups(P<0.05),and the effect of Jianpi Jiedu Decoction group was the best.In drug intervention group,the expression of IL-17,IFN-γ,IL-6 was significantly higher then that of blank control group,spleen deficiency model group and spleen deficiency psoriasis group(P<0.05),but significantly lower then that of spleen deficiency model group and spleen deficiency psoriasis group(P<0.05).Obvious psoriasis occurred in spleen deficiency psoriasis group.All treatment groups had different degrees of remission,and the Jianpi Jiedu Decoction group was better than other treatment groups.Conclusion:Jianpi Jiedu Decoction can regulate the expression levels of IFN-γ,IL-6 and IL-17,it is providing more sufficient scientific basis for establishing a new target of psorasis treated with traditional Chinese medicine compound.
