摘要
目的:建立HPLC-DAD法以定量分析黄芪-红花药对水提液中羟基红花黄色素A、紫丁香苷、毛蕊异黄酮苷、脱水红花黄色素B、山柰酚-3-O-芸香糖苷、芒柄花苷、毛蕊异黄酮、黄芪甲苷8种有效成分。方法:色谱柱为Alltech Alltima C18(250 mm×4.6 mm,5μm),流动相为乙腈-0.01%三氟乙酸,梯度洗脱,柱温30℃,流速1.0 mL/min,检测波长210 nm(毛蕊异黄酮苷、黄芪甲苷)、260 nm(紫丁香苷、山柰酚-3-O-芸香糖苷、芒柄花苷、毛蕊异黄酮)、403 nm(羟基红花黄色素A、脱水红花黄色素B)。结果:8种成分的浓度与峰面积间均呈现良好线性关系(r≥0.9997),精密度、稳定性和重复性良好,平均加样回收率为96.27%~101.31%,RSD为0.74%~2.79%。结论:该方法适用于黄芪-红花药对水提液中8种有效成分含量的同时测定,可用于黄芪-红花药对水提液的质量控制。
Objective:To establish an HPLC-DAD method for quantitative determination of hydroxysafflor yellow A,syringin,calycosin-7-O-β-D-glycoside,anhydrosafflor yellow B,kaempferol-3-O-rutinoside,ononin,calycosin and astragaloside Ⅳ in aqueous extract solution of Astragali Radix-Carthami Flos drug pair.Methods:HPLC-DAD experiment was performed on an Alltech Alltima C18(250 mm×4.6 mm,5 μm) column with mobile phase of acetonitrile-0.01% trifluoroacetic acid aqueous solution in a gradient elution manner.The column temperature and flow rate of the mobile phase were set at 30 ℃ and 1.0 mL/min.The detection wavelength was 210 nm for calycosin-7-O-β-D-glycoside,astragaloside Ⅳ 260 nm for syringin,kaempferol-3-O-rutinoside,ononin,calycosin,and 403 nm for hydroxysafflor yellow A,anhydrosafflor yellow B.Results:The aforesaid eight ingredients showed benign linearity(r≥0.9997),precision,stability and repeatability.The average recoveries were among 96.27%-101.31% with RSD of 0.74%-2.79%.Conclusion:The method is suitable for the determination of eight ingredients in Astragali Radix-Carthami Flos drug pair,and can be applied to the quality control of the aqueous extract Astragali Radix-Carthami Flos drug pair.
