基于16S rRNA测序分析陈皮-枳壳对创伤后应激障碍大鼠肠道菌群的影响 被引量:6

Investigate the structural and functional changes of intestinal flora in PTSD rats after treatment with tangerine peel and fructus aurantii by 16S rRNA sequence technique

摘要 目的:通过16S rRNA测序技术探究创伤后应激障碍(PTSD)大鼠经陈皮-枳壳治疗后肠道菌群结构及功能变化。方法:将24只SD大鼠随机分为正常组、模型组和治疗组,每组8只。采用单次延长应激法复制PTSD模型,治疗组给予0.5 g·kg^(-1)·d^(-1)陈皮-枳壳颗粒剂混合溶液,其余两组给予等量蒸馏水。7 d后收集粪便样本,运用16S rRNA测序技术与生物信息学分析方法得出结论。行为学测试包括旷场实验和僵立行为测试。结果:与模型组比较,治疗组直立次数显著增加(P<0.01),僵立时间显著减少(P<0.01);拟杆菌门各水平差异菌群相对丰度上调,厚壁菌门、变形菌门和蓝藻菌门各水平差异菌群相对丰度下调。结论:陈皮-枳壳可能是通过改变肠道菌群环境,促进拟杆菌门相关细菌繁殖,抑制厚壁菌门、变形菌门和蓝藻菌门部分细菌生长,这是其治疗PTSD可能的生物学机制。 Objective:To investigate the structural and functional changes of intestinal flora in post-traumatic stress disorder(PTSD)rats after treatment with tangerine peel and fructus aurantii by 16S rRNA sequence technique.Methods:A total of 24 SD rats were randomly divided into normal group,model group and treatment group,with 8 rats in each group.SPS method was used to replicate the PTSD model.Rats in the treatment group were given a solution of tangerine peel and fructus aurantii at0.5 g·kg^(-1)·d^(-1).Fecal samples were collected 7 days later,and the conclusions were drawn by using 16S rRNA sequencing technology and bioinformatics analysis.Behaviour texts include the open?eld test and freezing time text.Results:Compared with model group,the number of standing upright was signi?cantly increased(P<0.01)and the freezing time was signi?cantly decreased after treatment(P<0.01),meanwhile,the relative abundance of the dominant bacteria of Bacteroidetes increased at different levels,while the relative abundance of Firmicutes,Proteobacteria and Cyanobacteria decreased at different levels.Conclusion:Tangerine peel and fructus aurantii may change the environment of intestinal?ora,so that the Bacteroidetes related bacteria can promote reproduction,some related bacteria of Firmicides,Proteobacteria and Cyanobacteria are inhibited.This is the possible biological mechanism of its treatment of PTSD.
