FOXP3、MUC5AC在支气管炎患儿中的表达及与Th9细胞因子的相关性研究 被引量:2

摘要 目的探讨叉状头/翼状螺旋转录因子(FOXP3)、黏蛋白5AC(MUC5AC)在支气管炎患儿中的表达及与辅助性T细胞9(Th9)细胞因子的相关性。方法选取2018年2月至2021年2月该院支气管炎患儿94例作为观察组,选取同期健康体检儿童45例作为对照组。采用逆转录聚合酶链反应(RT-PCR)测定FOXP3 mRNA、MUC5AC mRNA水平,采用流式细胞学检测Th9细胞,采用酶联免疫吸附法(ELISA)检测白细胞介素-9(IL-9)水平,比较两组FOXP3 mRNA、MUC5AC mRNA、IL-9水平及Th9细胞比例,分析支气管炎患儿FOXP3 mRNA、MUC5AC mRNA及Th9细胞比例与病情严重程度的相关性,FOXP3 mRNA、MUC5AC mRNA与Th9细胞因子的相关性,Logistic回归分析支气管炎患儿预后的影响因素。结果观察组FOXP3 mRNA水平低于对照组,MUC5AC mRNA、IL-9水平及Th9细胞比例高于对照组,差异有统计学意义(P<0.05);支气管炎患儿FOXP3 mRNA与病情严重程度呈负相关,MUC5AC mRNA、Th9细胞比例、IL-9与病情严重程度呈正相关(P<0.05);支气管炎患儿FOXP3 mRNA与Th9细胞比例、IL-9呈负相关,MUC5AC mRNA与Th9细胞比例、IL-9呈正相关(P<0.05);Logistic回归模型分析显示,将体质、病情程度控制后,FOXP3 mRNA低表达及FOXP3 mRNA、IL-9高表达及Th9细胞比例升高为喘息发作危险因素(P<0.05)。结论支气管炎患儿FOXP3 mRNA、MUC5AC mRNA均异常表达,可能是Th9细胞因子高表达的重要因素,共同参与小儿支气管炎发病及病情进展,且各指标异常表达均为喘息发作危险因素。 Objective To investigate the expression of fork head/wing spiral transcription factor(FOXP3)and Mucin 5 AC(MUC5 AC)in children with bronchitis and their correlation with helper T cell 9(Th9)cytokines.Methods From February 2018 to February 2021,a total of 94 children with bronchitis in the hospital were selected as the observation group,and 45 healthy children who underwent the healthy physical examination were selected as the control group.Reverse transcription polymerase chain reaction(RT-PCR)was used to detect the levels of FOXP3 mRNA and MUC5 AC mRNA,flow cytometry was used to detect Th9 cells,and enzyme-linked immunosorbent assay(ELISA)was used to detect the level of interleukin-9(IL-9).FOXP3 mRNA,MUC5 AC mRNA,IL-9 levels and the Th9 cell ratio were compared between the two groups.The correlation between FOXP3 mRNA,MUC5 AC mRNA and Th9 cyto cell ratio in children with bronchitis and the severity of disease,and the correlation between FOXP3 mRNA,MUC5 AC mRNA and Th9 cyto cell ratio were analyzed.Logistic regression was used to analyze the factors affecting the prognosis of children with bronchitis.Results FOXP3 mRNA level in the observation group was lower than that in the control group,and MUC5 AC mRNA and IL-9 levels and Th9 cell ratio were higher than those in the control group,and the differences were statistically significant(P<0.05).FOXP3 mRNA in children with bronchitis was negatively correlated with the severity of the disease,and MUC5 AC mRNA,Th9 cell ratio,and IL-9 were positively correlated with the severity of the disease(P<0.05).In children with bronchitis,FOXP3 mRNA was negatively correlated with Th9 cell ratio and IL-9,and MUC5 AC mRNA was positively correlated with Th9 cell ratio and IL-9(P<0.05).Logistic regression model analysis showed that after controlling the physique and disease severity,low expression of FOXP3 mRNA,high expression of FOXP3 mRNA and of IL-9 and in crease of Th9 cell ratio were risk factors for wheezing onset(P<0.05).Conclusion Both FOXP3 mRNA and MUC5 AC mRNA are abnormally expressed in children with bronchitis,which may be important factors in the high expression of Th9 cyto cell.They are jointly involved in the onset and progression of bronchitis in children,and the abnormal expression of each index is a risk factor for wheezing onset.
