摘要
目的 研究玉屏风散治疗慢性湿疹的作用机制。方法 采用网络药理学研究方法,运用中药系统药理数据库和分析平台(TCMSP)筛选玉屏风散的有效成分及其作用靶点。通过GeneCards数据库筛选慢性湿疹的靶点,使用STRING平台对二者交集的靶点蛋白构建蛋白相互作用网络,并筛选关键靶点。使用DAVID 6.8工具对交集靶点进行基因功能注释(GO)生物过程和京都基因组百科全书(KEGG)信号通路富集分析,探讨交集靶点可能有的生物学功能及其发生作用的通路。结果 从玉屏风散中筛选出44个化合物,涉及治疗慢性湿疹的83个靶点,网络分析结果显示该方可能通过细胞因子受体结合、细胞因子活性、血红素结合、四吡咯结合、受体配体活性、核受体活性、转录因子活性,直接的配体调节序列特异性脱氧核糖核酸(DNA)结合、磷酸酶结合、蛋白磷酸酶结合、辅因子结合等生物过程发挥作用;其作用机制与糖尿病并发症中的晚期糖基化终末产物-糖基化终末产物受体(AGE-RAGE)信号通路、流体剪切应力与动脉粥样硬化、肿瘤坏死因子(TNF)信号通路、白细胞介素-17(IL-17)信号通路、恰加斯病(美国锥虫病)、甲型流感、卡波西氏肉瘤相关疱疹病毒感染、蛋白聚糖、C型凝集素受体信号通路、松弛素信号通路等有关。结论 玉屏风散治疗慢性湿疹有多成分、多靶点、多通路的特点,其作用机制主要集中在对炎性反应、免疫反应相关通路的调控。
Objective In order to study the mechanism of Yupingfeng powder in treating chronic eczema. Methods The network pharmacology research methods and the Chinese medicine system pharmacology database and analysis platform(TCMSP) were used to screen the effective components of Yupingfeng powder and its targets. The target of chronic eczema was screened through the Genecards database,and the STRING platform was used to construct a protein interaction network for the target proteins that the two intersect,and then the key targets were screened. The DAVID 6.8 tool was used to analyze GO biological processes and Kyoto Encyclopedia of Genes and Genomes(KEGG) signal pathway enrichment on intersection targets,to explore the possible biological functions and pathways of intersection targets. Results Forty-four compounds were screened from Yupingfeng powder,involved 83 targets for the treatment of chronic eczema. The results of network analysis showed that the compound may be bound by cytokine receptor binding,cytokine activity,heme binding,tetrapyrrole binding,Ligand activity,nuclear receptor activity,direct ligand regulation sequence-specific deoxyribonucleic acid(DNA) binding phosphatase binding,protein phosphatase binding,cofactor binding and other biological processes play a role;its mechanism of action was related to diabetes complications AGE-RAGE signaling pathway,fluid shear stress and atherosclerosis,tumor necrosis factor(TNF) signaling pathway,IL-17 signaling pathway,Chagas disease(American trypanosomiasis),influenza A,Kaposi’s sarcoma-related herpes virus infection,proteoglycan in cancer,C-type lectin receptor signaling pathway,relaxin signaling pathway and other signaling pathways. Conclusion The treatment of chronic eczema with Yupingfeng powder has the characteristics of multiple components,multiple targets,and multiple pathways,and its mechanism of action is mainly concentrated on the regulation of pathways related to inflammation and immune response.
