线粒体靶向抗氧化剂SS31对家兔卵巢组织冷冻效果的影响研究

摘要 目的 探讨线粒体靶向抗氧化剂SS31(D-Arg-2,6-dimethyltyrosine-Lys-Phe-NH2)对家兔卵巢组织冷冻效果的影响。方法 将15只家兔去势,获得30个卵巢,随机分成FRESH组(新鲜卵巢皮质片段)、CONTROL组(常规冷冻复苏,卵巢冷冻保护液和复苏液中无SS31)、5μM-SS31组、10μM-SS31组、20μM-SS31组、40μM-SS31组:在卵巢冷冻保护液和复苏液中分别加入5、10、20、40μM SS31(n=5)。分析比较各组中各级卵泡形态、数量、卵泡损伤凋亡情况、卵巢超微结构的改变。结果 与FRESH组比较,冻融各组卵泡数量减少、大量卵泡损伤凋亡,细胞器不同程度形态改变。随着SS31浓度的增加,冻融组组织学形态改善,卵泡损伤率和凋亡率降低,原始卵泡与初级卵泡数量增加。其中40μM-SS31组效果显著(P<0.05)。结论 SS31可减轻冻融卵巢组织中卵泡的损伤,对于玻璃化冷冻和复苏过程具有一定的保护作用。 Objective To investigate the effect of mitochondrial targeted antioxidant SS31(D-ARg-2,6-dimethyltyrosine-Lys-Phe-NH2) on the freezing effect of rabbit ovarian tissue.Methods Fifteen rabbits were selected and 30 ovaries were obtained, which were randomly divided into FRESH group(fresh ovarian cortical fragment),CONTROL group: routine cryopreservation, no SS31 in ovarian cryoprotective solution and resuscitation solution;5 μM-SS31 group, 10 μM-SS31 group, 20 μM-SS31 group, 40 μM-SS31 group: Add 5,10,20,40 μM SS31 to ovarian cryoprotective solution and resuscitation solution, respectively(n=5).The morphology and quantity of follicles at all levels, the damage and apoptosis of follicles, and the changes of ovarian ultrastructure in each group were analyzed and compared.Results Compared with the FRESH group, the number of follicles decreased, a large number of follicles were damaged and apoptotic, and the morphology of organelles changed to varying degrees in all freeze-thaw groups.With the increase of SS31 concentration, the histological morphology of freezing-thawing group was improved, the follicle damage rate and apoptosis rate were decreased, and the number of primordial and primary follicles was increased.The effect of 40 μM-SS31 group was significant(P<0.05).Conclusion SS31 can reduce the damage of follicles in freezing-thawing ovarian tissue, and has a certain protective effect on vitrification freezing and resuscitation.
