淫羊藿苷对肺炎支原体肺炎小鼠炎症和Notch信号影响 被引量:2

Effect of icariin on inflammation and Notch signal in mice with Mycoplasma pneumoniae pneumonia

摘要 目的研究淫羊藿苷对肺炎支原体肺炎小鼠炎症和Notch信号影响。方法实验动物分成空白组、模型组、低剂量实验组(100 mg·kg^(-1)淫羊藿苷灌胃)、高剂量实验组(200 mg·kg^(-1)淫羊藿苷灌胃)、对照组(阿奇霉素灌胃)。取肺组织,测量肺湿重指数,用苏木精-伊红(HE)染色评价肺组织病理学评分,用酶联免疫吸附(ELISA)法检测白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)含量,用黄嘌呤氧化法测定超氧化物歧化酶(SOD)含量,用硫代巴比妥酸法测定丙二醛(MDA)含量,用Griess法检测一氧化氮(NO)含量,用蛋白质印迹(Western blot)法测定诱导型一氧化氮合酶(iNOS)、Notch1、HES1、HEY1蛋白表达。结果空白组、模型组、对照组和低、高剂量实验组小鼠肺湿重指数分别为0.74±0.07,0.93±0.09,0.77±0.08,0.85±0.06和0.78±0.07,肺组织病理评分分别为5.50±1.47,14.58±1.84,7.95±1.35,10.29±1.74和8.32±1.87,IL-6含量为(90.26±8.15),(140.53±12.25),(115.84±12.53),(123.01±11.47)和(111.58±10.69)ng·L^(-1),IL-1β含量为(60.17±5.22),(113.74±12.98),(80.79±7.14),(96.67±10.44)和(82.45±8.76)ng·L^(-1),TNF-α含量为(347.12±33.17),(657.49±75.23),(375.49±40.97),(512.24±67.81)和(401.72±46.32)ng·L^(-1),IL-8含量为(410.36±55.29),(795.32±71.44),(520.23±45.91),(634.12±81.69)和(514.79±66.28)ng·L^(-1),SOD含量为(61.23±7.51),(16.84±2.56),(34.26±4.59),(26.58±2.45)和(36.69±4.78)U·mL^(-1),肺组织MDA含量分别为(25.36±2.81),(108.95±15.27),(59.95±8.72),(73.54±10.52)和(60.72±6.54)μmol·mL^(-1),肺组织NO含量分别为(201.47±16.28),(654.21±67.84),(478.35±57.82),(527.86±73.02)和(462.29±53.18)μmol·L^(-1);低剂量实验组、高剂量实验组、对照组与模型组比较,差异均有统计学意义(均P<0.05);高剂量实验组与低剂量实验组比较,差异均有统计学意义(均P<0.05)。结论淫羊藿苷可能通过抑制Notch信号减少NO合成和氧化应激,进而抑制肺组织炎症。 Objective To study that effect of icariin on inflammation and Notch signal in mice with Mycoplasma pneumoniae pneumonia.Methods The experimental animals were divided into normal group,model group,low dose experimental group(100 mg·kg^(-1) icariin by gavage),high dose experimental group(200 mg·kg^(-1) icariin by gavage),and control group(azithromycin by gavage).The lung tissue was taken and the lung wet weight index was measured.The pathological score of the lung tissue was evaluated by hematoxylin-eosin(HE)staining.The contents of interleukin-6(IL-6),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)and interleukin-8(IL-8)were detected by enzyme-linked immunosorbent assay(ELISA);the content of superoxide dismutase(SOD)was determined by xanthine oxidation method;the content of malondialdehyde(MDA)was determined by thiobarbituric acid method;the content of nitric oxide(NO)was determined by Griess method;the protein expressions of inducible nitric oxide synthase(iN OS),Notch1,HES1 and HEY1 were determined by Western blot.Results The lung wet weight indexes of normal group,model group,control group and low,high dose experimental groups were 0.74±0.07,0.93±0.09,0.77±0.08,0.85±0.06 and 0.78±0.07;the lung tissue pathological scores were 5.50±1.47,14.58±1.84,7.95±1.35,10.29±1.74 and 8.32±1.87;IL-6 content were(90.26±8.15),(140.53±12.25),(115.84±12.53),(123.01±11.47)and(111.58±10.69)ng·L^(-1);IL-1β content were(60.17±5.22),(113.74±12.98),(80.79±7.14),(96.67±10.44)and(82.45±8.76)ng·L^(-1);TNF-α content were(347.12±33.17),(657.49±75.23),(375.49±40.97),(512.24±67.81)and(401.72±46.32)ng·L^(-1);IL-8 content were(410.36±55.29),(795.32±71.44),(520.23±45.91),(634.12±81.69)and(514.79±66.28)ng·L^(-1);SOD content were(61.23±7.51),(16.84±2.56),(34.26±4.59),(26.58±2.45)and(36.69±4.78)U·mL^(-1);MDA content in lung tissue were(25.36±2.81),(108.95±55.27),(59.95±8.72),(73.54±10.52)and(60.72±6.54)μmol·mL^(-1);NO content in lung tissue were(201.47±16.28),(654.21±67.84),(478.35±57.82),(527.86±73.02)and(462.29±53.18)μmol·L^(-1).Compared with the differences between model group and low dose experimental group,high dose experimental group,control group were statistically significant(all P<0.05);the difference between high dose experimental group and low dose experimental group were statistically significant(all P<0.05).Conclusion Icariin may reduce NO synthesis and oxidative stress by inhibiting Notch signaling,thereby inhibiting inflammation in lung tissue.
