肾上腺皮质癌中CENPF基因的表达及对患者治疗和预后的影响

Expression of CENPE gene in adrenocortical carcinoma and its influence on treatment and prognosis of patients

目的 探讨着丝粒蛋白F(centromere protein F,CENPF)在肾上腺皮质癌(adrenocortical carcinoma,ACC)中的表达及临床意义。方法 采用免疫组化EnVision两步法检测ACC组织中CENPF蛋白的表达,并结合GEO数据库、GEPIA平台中CENPF mRNA的表达信息,对CENPF在ACC中的表达及临床意义进行分析,绘制患者生存曲线。应用RNA干扰实验分析CENPF在ACC细胞系SW13中的潜在生物学行为。结果 在临床组织及肿瘤相关数据库中,与正常肾上腺皮质组织及肾上腺皮质腺瘤相比,CENPF在ACC中呈高表达(P<0.05)。CENPF表达量与ACC TNM分期呈显著正相关(P=0.000 174),与ACC患者的总生存期、无进展生存期呈显著负相关(HR=8.88、4.12,P均<0.001),且CENPF可能是ACC患者的独立预后因素(P<0.05)。RNA干扰实验表明,下调CENPF表达可抑制SW13细胞的增殖、迁移能力(P均<0.05)。结论 CENPF在ACC组织中过表达,且与患者预后呈负相关,CENPF可能参与ACC的发生、发展。

Purpose To investigate the expression and clinical significance of centromere protein F(CENPF) in adrenocortical carcinoma(ACC).Methods The expression of CENPF protein in ACC tissues was detected by immunohistochemistry assay,and the expression of CENPF mRNA in GEO database and GEPIA platform was applied to analyze the clinical significance of CENPF in ACC,the patient survival curve was drawn.RNA interference experiment was applied to explore the potential biological function of CENPF in ACC.Results In clinical samples and tumor related databases,CENPF was significantly overexpressed in ACC compared with normal adrenal cortex and adrenocortical adenoma(P<0.05).The expression of CENPF was significantly positively correlated with the clinical stage of ACC(P=0.000 174),and negatively correlated with the prognosis of ACC patients,including overall survival and progression free survival(HR=8.88 and 4.12,all P<0.001).CENPF may serve as an independent prognostic factor of ACC patients.RNA interference assay showed that the downregulation of CENPF inhibited the proliferation and migration of SW13 cells(P<0.05).Conclusion The CENPF in ACC tissues is significantly overexpressed,and is associated with dismal prognosis in ACC patients,which suggested that CENPF may be involved in the occurrence and development of ACC.
