姜黄素对溃疡性结肠炎小鼠肠道损伤和免疫功能影响的机制研究 被引量:4

摘要 目的探讨姜黄素对溃疡性结肠炎小鼠肠道炎症、肠道屏障和免疫功能的影响并研究其可能的作用机制。方法随机将BALB/c小鼠分为四组,分别为对照组、模型组、姜黄素组和模型^(+)姜黄素组。模型组及模型^(+)姜黄素组采用3%葡聚糖硫酸钠(Dextran Sulfate Sodium,DSS)诱导溃疡性结肠炎模型。姜黄素组及模型^(+)姜黄素组用50 mg/kg姜黄素(5%环糊精溶解)进行干预,连续7天,每日观察小鼠的体重变化和便血程度。采用RT-PCR技术检测小鼠结肠组织炎症因子和肠道屏障相关因子的表达,流式细胞术检测小鼠肠系膜淋巴结CD3^(+)CD4^(+)T细胞和CD3^(+)CD8^(+)T细胞的变化。结果与模型组相比,模型^(+)姜黄素组小鼠体重增加,结肠长度增长,结肠组织炎症因子NLRP3、IL-6、TNF-α和COX-2的mRNA表达水平降低,肠道屏障相关因子MMP-9的mRNA表达水平降低,MUC-2和ZO-1的mRNA表达水平增加以及肠系膜淋巴结中CD3^(+)CD4^(+)T细胞显著增加,CD3^(+)CD8^(+)T细胞显著减少。结论姜黄素可以抑制DSS诱导的炎症反应,保护肠道屏障功能,调控肠系膜淋巴结T细胞的平衡,从而达到治疗溃疡性结肠炎的目的。
