Quality evaluation of Rhizoma Sparganii based on phenolic compounds

摘要 目的 研究三棱中8种酚醛类成分在块茎外皮、内部以及根部和叶柄残基中的分布以及熏硫对这些成分的影响,为三棱药材的采收加工、贮藏提供科学依据。方法 采用高效液相色谱法(HPLC)测定样品中5-羟甲基糠醛、香兰素、原儿茶酸、香草酸、阿魏酸、咖啡酸、对香豆酸、1-O-阿魏酸-3-对香豆酸甘油酯的含量。结果 除5-羟甲基糠醛、香草酸外,外皮和叶柄残基中酚醛类成分含量均高于块茎内部;熏硫使5-羟甲基糠醛的含量增加,与熏制时间呈正相关;对香豆酸、1-0-阿魏酸-3-对香豆酸甘油酯含量在熏硫12h后明显降低。结论 高效液相法同时测定8种酚酸类成分可以用于评价三棱药材质量;采制过程中,去除须根和叶柄,削去外皮的加工方法有待商榷;熏硫对三棱品质有显著影响,在采收过程中不得为了便于干燥采用熏硫的措施,贮藏过程中不宜采用熏硫的方法杀虫。 Objective To study the distribution of 8 phenolic components in the tuber skin,interior,root and petiole residues and the effect of sulfur smoking on these components in the trigonatum,so as to provide a scientific basis for the harvesting,processing and storage of the herbs of Rhizoma Sparganii.Methods Determination of 5-hydroxymethylfurfural,vanillin,protocatechuic acid,coumaric acid,ferulic acid,caffeic acid,p-coumaric acid and 1-O-ferulic acid-3-p-coumaric acid glycerides in the samples by HPLC.Results Except for 5-hydroxymethylfurfural and vanillic acid,the contents of phenolic compounds in pericarp and petiole residues were higher than those in tuber.The content of 5-hydroxymethylfurfural was increased by sulfur fumigation,which was positively correlated with the fumigation time.The contents of p-coumaric acid and 1-0-ferulic acid-3-p-coumaric acid triglyceride decreased significantly after 12 h of sulfur fumigation.Conclusion Simultaneous determination of 8 phenolic acids by HPLC can be used to evaluate the quality of?Rhizoma Sparganii.The processing method of removing fibrous roots and petioles and peeling off skin in the process of harvesting remains to be discussed.Sulfur fumigation has a significant effect on the quality of Rhizoma Sparganii,so sulfur fumigation should not be used in the harvesting process to facilitate drying,and sulfur fumigation should not be used in the storage process to kill insects.
