摘要
试验旨在探究RPL38基因对狮头鹅肌肉发育进程的影响。选取15胚龄狮头鹅胚胎,分离出腿肌中的成肌细胞,并进行相关分子试验验证。采用RACE试验扩增出狮头鹅肌肉组织RPL38基因的全长mRNA,并参考mRNA序列克隆出表达载体。利用细胞周期检测试验和EDU增殖试验检测成肌细胞增殖,利用qRT-PCR检测肌肉分化相关基因的表达。结果发现RPL38蛋白的进化保守程度极高,但mRNA序列则保守程度很低。RPL38基因在狮头鹅成肌细胞增殖期高表达,而在分化中表达显著下调,通过在狮头鹅成肌细胞中过表达和干扰RPL38基因,发现RPL38会促进成肌细胞的增殖,但不影响成肌细胞分化。结果表明,RPL38可通过调控鹅成肌细胞增殖,影响鹅肌肉的生长发育。
The aim of this study was to investigate the effects of RPL38 gene on the muscle development of Shitou geese. 15-day-incubated Shitou goose embryos were selected to isolate the myoblasts in the leg muscles for related molecular experiments.The RACE experiment was used to evaluate the full-length mRNA of the RPL38 gene in Shitou goose muscle tissue, and the RPL38 over expression vector was cloned with reference to the mRNA sequence. Cell cycle detection experiment and EDU proliferation experiment were used to detect the proliferation of myoblasts, and qRT-PCR was used to detect the expression of genes related to muscle differentiation. It was found that RPL38 protein was highly conserved among vertebrates, but the mRNA sequence was not. The expression of RPL38 gene was significantly down regulated during myoblast differentiation of Shitou goose. Ectopic expression of RPL38 in goose myoblast indicated that RPL38 could promote myoblast proliferation but had no significant effect on myoblast differentiation. These results indicated that RPL38 might affect goose skeletal muscle growth and development via regulate myoblast proliferation.
