高效液相色谱法测定浓香窖泥中乳酸含量

摘要 建立了采用高效液相快速准确的检测窖泥中乳酸的含量的方法。此方法特征在于在样品前处理过程中加入磁力搅拌能很大程度的减少因窖泥结团而导致浸泡不均匀。由于窖泥中物质比较复杂,因此采用减压蒸馏的方法来处理样品能祛除大部分低沸点酯类化合物杂质,从而能使乳酸更好的分离。采用Agilent Polaris C18-A(4.6 mm×250 mm,5μm)色谱柱,以甲醇和磷酸二氢钾为流动相,体积比为5∶95,流速为0.5 mL/min,检测波长为210 nm。在此色谱条件下,乳酸在标准品和样品中出峰时间一致(出峰时间18.5 min),峰型较好,且加标回收率达到了98.7%~101.93%。 A rapid and accurate method for detecting the content of lactic acid in pit mud by high performance liquid phase was established.This method is characterized in that the addition of magnetic stirring during the pretreatment of the sample can greatly reduce the uneven soaking caused by the agglomeration of the pit mud.Since the substances in the pit mud are relatively complex,the vacuum distillation method can be used to treat the sample to remove the Most of the low-boiling esters are impurities,so that the lactic acid can be separated better.An Agilent Polaris C18-A(4.6 mm×250 mm,5 μm) chromatographic column was used,methanol and potassium dihydrogen phosphate were used as mobile phases,the volume ratio was 5:95,the flow rate was 0.5 mL/min,and the detection wavelength was 210 nm.Under this chromatographic condition,the peak time of lactic acid in the standard product and the sample is the same(peak time 18.5 min),the peak shape is good,and the recovery rate of standard addition reaches 98.7%~101.93%,and the RSD% value is 0.07~0.22.
