摘要
目的比较近端与远端胃癌患者胃内微生态结构和多样性差异。方法选择胃癌患者83例,其近端胃癌患者55例,远端胃癌患者28例。应用细菌16SrRNA基因扩增子测序技术,分析所有患者胃内微生态结构与多样性。结果两组样本经过滤后平均共获得序列30256条,用于构建OTUs,取得分类Tags数平均为24201条。两组患者总数Tags、分类单元tags、Total OTUS数目、Species OTUS数目、Genus OTUS数目、Species Chao 1、Genus Chao 1、Species Shannon指数比较均无显著差异(P>0.05),近端胃癌组Genus Shannon指数水平高于远端胃癌组(P<0.05);近端胃癌组胃内相对丰度前3位菌门依次为:变形菌门为57.10%,泉古菌门为14.27%,拟杆菌门为8.29%。远端胃癌组前3位菌门依次为:变形菌门为56.42%,泉古菌门为14.11%,拟杆菌门为8.12%。两组胃内细菌群落中,变形菌门丰度比例最高。近端胃癌相对丰度前3菌属依次为:不动杆菌属占5.22%,斯瓦尼菌属占3.74%,普里沃菌属占2.43%;远端胃癌相对丰度前3菌属:不动杆菌属占5.34%,斯瓦尼菌属占3.73%,普里沃菌属占2.41%;两组胃内细菌属水平构成无明显差异(P>0.05)。近端、远端胃癌组胃内相关细菌群落构成的PCoA分析显示,横坐标PC1变化率解释为12.83%,纵坐标PC2变化率解释为8.01%;经PCoA分析无法将近端、远端胃癌组相关细菌群落结构有效区分。结论近端与远端胃癌患者胃内微生态结构与多样性无明显差异,其相似性较高。
Objective To investigate the differences of gastric micro-ecological structure and diversity of patients with proximal and distal gastric cancer.Methods 83 gastric cancer patients were enrolled,including 55 patients with proximal gastric cancer and 28 patients with distal gastric cancer.The gastric micro-ecological structure and diversity of all patients were analyzed by 16SrRNA gene amplicon sequencing technology.Results After filtering,the 2 groups of samples obtained an average of 30256 sequences for constructing OTUs,and the average number of classified Tags obtained was 24201.Total number of patients in the 2 groups Tags,taxon Tags,Total OTUS number,Species OTUS number,Genus OTUS number,Species Chao 1,Genus Chao 1,Species There was no significant difference in Shannon index(P>0.05).The Genus Shannon index level in proximal gastric cancer group was higher than that in distal gastric cancer group(P<0.05).In the proximal gastric cancer group,the top 3 bacteria in relative abundance were Proteobacteria(57.10%),Archaea(14.27%)and Bacteroidetes(8.29%).The top 3 bacteria in distal gastric cancer group were Proteobacteria 56.42%,Archaea 14.11%,Bacteroidetes 8.12%.Proteobacteria abundance ratio was the highest in the two groups.The top 3 bacteria in relative abundance of proximal gastric cancer were Acinetobacter(5.22%),Swaniella(3.74%)and Privotella(2.43%).The top 3 genera in relative abundance of distal gastric cancer were Acinetobacter(5.34%),Swaniella(3.73%)and Privotella(2.41%).There was no significant difference in the composition of bacterial genus level between the two groups(P>0.05).PCoA analysis of related bacterial community composition in the stomach of proximal and distal gastric cancer groups showed that the change rate of PC1 in abscissa was 12.83%,and that of PC2 in ordinate was 8.01%.PCoA analysis could not effectively distinguish the bacterial community structure of proximal and distal gastric cancer groups.Conclusion There is no significant difference in the diversity of gastric microecological structure between proximal and distal gastric cancer patients.
